【Abstract】 AIM: To construct the eukaryotic expression plasmid pcDNA3/mIFNγ and observe its effect on secreting coxsackievirus B3(CVB3) VP1 gene vaccine. METHODS: mIFNγ cDNA amplified by RTPCR was inserted in eukaryotic expression vector pcDNA3 and then detected after being transfected into cos7 cells with DEAEDextran; Balb/c mice were inoculated in quadriceps muscle at 2week interval for 6 weeks with pcDNA3/sVP1, pcDNA3/sVP1+ pcDNA3/mIFNγ, respectively. The levels of serum antibodies were detected 13 d after each inoculation.The mice were challenged by 500 TCID50 CVB3 2 weeks after the last immunization. RESULTS: Recombinant pcDNA3/mIFNγ plasmid had been constructed and transcripted and expressed. The neutralizing antibody could be induced by pcDNA3/sVP1 and pcDNA3/sVP1+pcDNA3/mIFNγ; The antibody titers increased with the times of inoculation. But there was no difference in the antibody titers and the survival rate between the 2 groups. Coadministration of pcDNA3/mIFNγ and pcDNA3/sVP1 plasmid to Balb/c mice, decreased the titers of virus and reduced the histopathological changes compared with control groups. CONCLUSION: mIFNγ can enhance the immune protection of secreting CVB3 DNA vaccine. ......
