【Abstract】 AIM: To study the correlation between single nucleotide polymorphisms (SNPs)11377C/G and serum adiponectin level in the Han population with type 2 diabetes in Shaanxi province. METHODS: The PCR RFLP techniques were adopted to test SNPs in the proximal promoter region of the APM1 gene in 101 nondiabetes control group and 203 diabetes group. The diabetes patients underwent 75 g glucose OGTT and insulin release test for measurement of serum adiponectin, blood sugar, insulin and lipids. Insulin secretion index and insulin resistance index were calculated. RESULTS: Serum adiponectin level in the diabetes group were statistically lower than nondiabetes control group [(12.8±6.5) mg/L vs (15.0±6.9) mg/L, P<0.05]. The distribution of APM1 gene polymorphism in nondiabetes group and diabetes group was C/C53(52.5%), C/G38(37.6%), G/G10(9.9%) and C/C107(54.0%), C/G74(37.4%), G/G17(8.6%). In nondiabetes control group, serum adiponectin level of the GG genotype at position 11377 was lower than that of the CC genotype ,but the TG level of the GG genotype was much higher than that of the CC genotype (P<0.05). CONCLUSION: SNPs11377C/G modulate the adipocytesecreted adiponectin hormone level and may contribute to the genetic risk for type 2 diabetes in Shaanxi Chinese. ......
