【Abstract】 AIM: To observe the changes of the cell cycle and cytoskeleton of rat calvarial osteoblasts after stimulated by recombinant human growth hormone (rhgh ). METHODS: Osteoblastic cells from rats were incubated in the medium supplemented wiht rhgh at different doses (10, 200 μg/L), and then the changes of cytoskeleton and cell cycle were detected. RESULTS: Quantitative analysis showed that osteoblasts were arrested in S phase and G2/M phase, however, green fluorescent density and red fluorescent density (nuclei) were not significantly changed. The filamentous Factin distributed in bunches and was stained strongly and evenly. The outline of nucleus was clear. CONCLUSION: Rhgh exerts a direct anabolic effect on osteoblasts, but no obvious effect on cytoskeleton of rat osteoblasts. ......
