    Abstract

    AIM:
To investigate the changes of the Bcl-2 family in human gastric cancer cell line MKN 45 after epigallocatechin-3-gallate (EGCG) treatment.

    METHODS: The mRNA expression of apoptosis-related genes Bcl-xl, Bcl-2, Bax, Bid and Bad were examined by reverse transcription-polymerase chain reaction (RT-PCR). Western blot analysis was performed to detect the changes of the corresponding protein expression, and then the ratio of Bcl-2 to Bax (Bcl-2/Bax) was calculated.

    RESULTS: The mRNA and protein expression levels of pro-apoptotic members, such as Bax, Bid and Bad, were increased gradually as the treating time extended (Bax protein: 0.6292 ± 0.08, 0.6454 ± 0.04, 0.8069 ± 0.09, 0.8992 ± 0.07, P < 0.05; Bid protein: 1.1214 ± 0.08, 1.3697 ± 0.07, 1.610 ± 0.09, 2.0623 ± 0.12, P < 0.05; Bad protein: 0 ......
