【摘要】 目的 分析乙型肝炎病毒(hepatitis B virus, HBV)相关肝细胞癌(hepatocellular carcinoma, HCC)患者体内HBV X基因序列结构，判断HBV X基因变异与HCC发生有无相关性。方法 PCR法扩增HCC患者及其家族中HBV感染者血清HBV X基因，克隆并测序后对两组序列变异情况进行比较。结果 肝癌患者来源的HBV在HBx蛋白的T细胞表位、反式激活区、核心启动子区变异发生率较高，但与对照组相比差异无显著性(P>0.05)。结论 HBV X基因变异可能与HCC发生没有相关性，有必要进一步加大样本研究。【关键词】 肝细胞癌；乙型肝炎病毒；X基因；聚合酶链反应；基因变异
A study about mutations of X gene of HBV isolated from patients with HBV-related HCC

【Abstract】 Objective To study the structures of X gene of hepatitis B virus (HBV) isolated from patients with hepatocellular carcinoma (HCC) and to find out whether there have relations between mutations of HBV X gene and hepatocarcinogenesis.Methods HBV X genes were amplified from HBV DNA extracted from serum of HCC patients with positive HBsAg and their family members with positive HBsAg by polymerase chain reaction (PCR) respectively. The subgenomic frames were subjected to cloning and sequencing. Sequence results were aligned by BLAST and analyzed statistically.Results HBV strains isolated from HCC have high-rate-mutations in T cell epitopes of HBx protein, transactivating domain of HBx and basal core promoter regions but have no statistical significance in contrast with that of control group (P>0.05).Conclusion The point mutations in HBV X region may not be associated with the development of HCC, but it's necessary to study further more cases. ......
