泡状棘球蚴感染纯系大鼠肝移植免疫耐受的实验研究

     摘要: 目的:探讨感染泡球蚴的大鼠原位肝移植(Orthotopic liver transplantation, OLT)后免疫系统的变化特点及其对移植物的影响。方法:分为3组:(1)A组为正常对照组(同系移植):健康(Lewis)LEW大鼠→健康LEW大鼠;(2)B组为对照组(非同系移植):健康Brown Norway(BN)大鼠→健康LEW大鼠;(3)C组为实验组:BN大鼠→LEW大鼠(感染泡球蚴后3个月)。术后LEW大鼠存活48 h判定为肝脏移植手术成功。每组于术后1、3、5、7 d 4个时间点各处死2只LEW大鼠,取血清及移植的肝脏、脾脏。每组留8只用于观察生存期。结果:A组、B组、C组受体存活时间平均为(54.13±16.62) d、(4.75±1.58) d、(15.50±3.93) d,差异有统计学意义(P<0.05)。病理结果示:(1)A组无排斥反应现象;(2)B组排斥反应随移植天数的增加逐渐加重,术后1、3、5、7 d分别出现0、1、2、3级排斥反应;(3)C组第7天出现1级排斥反应。结论:感染泡状棘球蚴后有利于同种异体移植物的存活;该模型为研究感染泡球的肝脏移植提供了良好的实验平台

    Study on immune tolerance on orthotopic liver transplantation in the inbred rat infected with Echinococcosus multicularis

    Abstract: Objectives: To investigate the immune characteristics of rats infected with Echinococcosus multicularis (E.m) and its effects on the graft by using BNtoLEW orthotopic liver transplantation (OLT) models. Methods: The rats were randomly divided into three groups as follows: Group A normal controls were produced by (OLT in the same strain) from LEWto LEW; Group B.strain controls were produced by (OLT in the different strain) form BNtohealth LEW; Group C: E.m stimulated group were produced by BNtoLEW rat with E.m infection for 3 months; The OLT procedure was considered as succesed model when the LEW rat can be survived over 48 h. The LEW serum, liver graft, spleen samples were collected in 1, 3, 5, 7 days after OLT respectively. Eight LEW rats in each group were remained for observation of their the survival days. Results: The mean survival times for each group were (54.13±16.62)d for group A, (4.75±1.58) d for group B, (15.50±3.93) d for group C, respectively. The survival time had statistic difference. The pathology results indicated: (1) there were no rejection in group A; (2) in group B, there appeared class1 rejection in the third, class 2 rejection in the fifth and class 3 rejection in the seventh days repetitively after OLT in rats. (3) There were no rejection before the first 5 days in group C, but it were increased to class 1 rejection repectively in the seventh day. Conclusion: E.m Infection was beneficial to the survive after OLT, The models could be considered of a good platform for further study on the liver transplantation field, especially in relation to Echinococcosus infection. ......
