大鼠肝癌发生过程中bFGF的表达及意义(pdf)

    【摘要】 目的 探讨碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)在肝癌发生过程中的表达及意义。方法 采用免疫组织化学ABC法对二乙基亚硝胺(DEN)诱发大鼠肝癌发生过程中碱性成纤维细胞生长因子表达情况进行观察。结果 DEN诱发大鼠肝癌发生率为100%。正常大鼠肝细胞bFGF表达为阴性，于诱癌第8周，肝小叶近门管区血管附近的少量肝细胞呈bFGF阳性表达，并可见门管区血管内皮细胞和成纤维细胞也呈bFGF阳性表达，随着诱癌进程的发展，bFGF阳性肝细胞的数量逐渐增多，至诱癌第18周，癌结节内大部分肝癌细胞均呈bFGF阳性表达。结论 bFGF与肝癌的发生和发展具有相关性。

    【Abstract】 Objective To investigate the expression and significance of basophilic fibroblast growth factor during hepatocarcinigenesis in the rats.Methods Hepatoma was induced in the rats by administration of diethylnitrosamine (DEN),the expression of basic fibroblast growth factor (bFGF) was detected with immunohistochemical technique.Results The incidence of hepatocarcinogenesis induced by DEN was 100%,the expression of bFGF was not detected in hepatocytes of normal rats, a few of bFGF positive hepatocytes were detected near the fibrous septums between hepatic lobules at 8th week after DEN treatment, and the positive expression of bFGF was also detected in the endothelial cells of blood vessels and the fibroblast,the number of bFGF positive hepatocytes increased along with the process of carcinogenesis, the majority of hepatocytes of basophilic nodules expressed bFGF at 18th week after DEN administration.Conclusion bFGF is associated closely with the development and progression of hepatic carcinoma. ......
