     摘要: 目的观察异十三基二胺(D79)的体内外抗肿瘤作用。方法应用锥虫蓝排染法、MTT法检测D79对多种体外培养肿瘤细胞的细胞毒作用;Bliss法观察D79急性毒性实验;在可耐受剂量下观察D79对小鼠移植性实体瘤U14、腹水瘤EAC、HAC的抑制率。结果 D79显著杀伤多种体外肿瘤细胞,药物作用48 h,IC50 1.24~3.36 μg/mL;D79的LD50为36.49 mg/kg(静脉注射)。D79抑制小鼠移植性实体瘤U14体内生长、提高移植性腹水瘤EAC、HAC小鼠生存时间的作用呈剂量依赖性。结论 D79有较强的体内外抗肿瘤作用。

    Antitumor Activity of Allotritridecyldiamidogen in vivo and in vitro

    ABSTRACT: Objective To observe the antitumor effect of allotritridecyldiamidogen(D79) in vivo and in vitro. Methods The cytotoxic effects of D79 on various tumor cells lines cultured in vitro were determined by trypan blue dye exclusion test, MTT method . Acute toxicity of D79 was evaluated by Bliss method: at a tolerable dose level, D79 was administrated to treat transplanted solid tumor U14 and ascetic tumors EAC and HAC. Tumor weight inhibition and life extension were observed. Results D79 induced significant growth arrest in various malignant tumor cell lines cultured in vitro. IC50 of these tumor cells exposed to the drug for 48 hours ranged from 1.24~3.36 μg/mL. LD50 of D79 was 36.49 mg/kg(mice, iv). D79 inhibit in vivo growth of implanted solid tumor U14 and prolonged live time implanted ascetic tumors EAC and HAC of mice in a dosedependment manner. Conclusion D79 had obvious antitumor activity in vivo and in vitro. ......
