雌/雄激素诱导去势大鼠前列腺增生过程中间质细胞成分的动态变化

     【摘要】 目的 在建立雌/雄激素诱导大鼠前列腺增生动物模型的基础上,研究前列腺增生过程中间质细胞成分的动态变化。方法 对雄性Wistar大鼠施去势手术21天后,注射浓度比例为1:100的雌/雄激素,于给药第0、3、7、10、14、17和21天取材。选定前列腺指数变化最明显的时间,考察HE染色的组织切片、Vimentin和α-smooth actin(SMA)免疫组化染色等指标。结果 去势大鼠注射雌/雄激素后,前列腺指数逐渐增加,以第3和10天增加的最为明显,17天后变化趋于平缓。对3、10和21天的组织进行组织学观察。病理切片显示,在第3和10天前列腺间质成分明显增多,21天相对减少。SMA免疫组化显示,去势大鼠给药后腺泡腔外平滑肌层逐渐减少,给药10天左右接近正常,而后,又逐渐增厚。Vimentin免疫组化显示,去势后给药第3天Vimentin阳性细胞明显增多,但随着给药时间延长,阳性细胞逐渐减少。结论 雌/雄激素诱导去势大鼠前列腺增生的过程中间质细胞数量和表型有明显的动态变化,为进一步研究前列腺增生过程中病理改变和分子机制提供有益的线索。

    Dynamic changes of stromal cells in the process of estrogen/androgen induced prostatic hyperplasia in castrated rats

    【Abstract】 Objective To investigate the dynamic changes of stromal cells in the process of estrogen/androgen induced prostatic hyperplasia (BPH) in castrated rats.Methods Twenty-one days after castration of male Wister rats, the rats were injected subcutaneously with estrogen/androgen at a ratio of 1:100. The rats were sacraficed at 0, 3, 7, 10, 14, 17, 21, 28 days later and the prostates were subjected to prostate index (PI) analysis, hematoxylin and eosin (H&E) staining, immunohistochemistry (IHC) staining for vimentin and α-smooth muscle actin (SMA).Results After injection with estrogen/androgen, the PI of the castrated rats were gradually increased, especially at day 3 and day 10, while PI hardly increased after day 17. H&E staining indicated that the stromal compartment was obviously increased at day 3 and day 10, while decreased after day 21. IHC staining for SMA showed that the smooth muscle cell (SMC) layer surrounding the prostate gland lumen gradually get thinned after injection, thickened as the sham-operated rats at day 10 and become more thicker thereafter. IHC staining for vimentin showed that after injection,vimentin positive cells were markedly increased at day 3, while PF decreased thereafter. Conclusion The quantity and phenotype of stromal cells obviously changes in the process of estrogen/androgen induced benign prostatic hyperplasia in castrated rats. It may mimic the naturally-occurred physiological changes in the progress of prostate development and pathogenesis of BPH. ......
