【摘要】 目的:观察纯CO2气体环境对卵巢癌细胞SKOV3生长及运动能力的影响。方法:将体外培养的卵巢癌SKOV3细胞悬液接种于96孔板,将培养板分别暴露于纯CO2环境1、2、3h,对照组放入常规细胞培养箱培养。纯CO2处理后24、48、72、96、120h,用四唑盐(MTT法)比色试验测定各组细胞的比色值,观察细胞生长情况,绘制细胞生长曲线,观察各组细胞倍增时间。采用划痕(Woundassay)试验测定各组细胞随机运动能力的变化。结果:卵巢癌细胞SKOV3暴露于纯CO2环境后第24、48、72小时 MTT值与对照组无显著性差异,第96小时后其MTT值明显高于对照组,其差异程度与暴露于纯CO2环境时间呈正相关。实验组细胞倍增时间较对照组缩短。各组细胞随机运动能力无显著性差异。结论:纯CO2气体环境对卵巢癌细胞SKOV3生长的影响与作用时间有关,卵巢癌细胞暴露于纯CO2环境短时间(1~2h)内,对细胞生长无明显影响,3h后对肿瘤细胞的生长有明显促进作用。纯CO2环境对肿瘤细胞的随机运动能力无明显影响。【关键词】 二氧化碳;肿瘤细胞;细胞培养;细胞生长;细胞运动

【Abstract】 Objective:To observe the effects of carbon dioxide on growth and movement of human ovarian cancer cells in vitro Such study will provide a primary evidence for the safety of CO2 pneumoperitoneum laparoscopy surgery in malignanciesMethods:Immortalized ovarian epithelial carcinoma cell (SKOV3 cell line) cultures were exposed to carbon dioxideCell growth was determined by spectrophotometric assay(MTT)Cell movement was determined with the use of WoundassayThe results were compared with control cells by paired test and liner regressions analysis.Results:The levels of MTT of carcinoma cells that were exposed to carbon dioxide had increased after 4 days in cultureThe increased cell growth had a linear relationship to the length of carbon dioxide exposure.The doubling time of carcinoma cells exposed to carbon dioxide was shorter than that in control cells.The number of migrating cells (SKOV3)into the wound surface were no significant difference to be found among four groups (P>005)Conclusions:Carbon dioxide exposure increases the growth of human ovarian carcinoma cells in vitroThere is no effect on movement of SKOV3 cell after exposed to carbon dioxide ......
