低氧预适应诱导C6细胞对氧葡萄糖剥夺的耐受

     [摘 要] 目的: 研究低氧预适应诱导神经胶质细胞瘤细胞(C6 Glioma Cells,简称C6细胞)对氧葡萄糖剥夺(OGD) 的耐受及其机制。 方法: C6细胞通过低氧(1%)预处理,在经受OGD打击后,观测其对细胞活力的影响。利用West- ern blot分析低氧预适应对C6细胞磷酸化-ERK(p-ERK)、缺氧诱导因子-1α(HIF-1α)和红细胞生成素(EPO)蛋白的 影响;通过PD98059和LY294002的使用,分析低氧诱导C6细胞预适应现象与MEK/ERK通路的关系。 结果: 低氧能 诱导细胞预适应的发生,而且对于24 h的OGD,最有效的低氧预适应时间为16 h;低氧能诱导细胞p-ERK信号蛋白 的上调,激活HIF-1α和增加EPO的表达,且能被PD98059所抑制。 结论: 低氧预适应能增强C6细胞对24 hOGD的 耐受,产生预保护作用,其中以低氧预适应16 h效率最高;MEK/ERK信号转导途径参与了低氧预适应作用;低氧预适 应可通过激活HIF-1α和增加EPO表达水平来实现。

    Hypoxia preconditioning induces tolerance against oxygen glucose deprivation in C6 glioma cells

Methods: The C6 cellular viability changes were observed after the pretreatment of hypoxia(1% O2) and the attack of OGD. Using Western blot, we analyzed the influence on the expression of p- ERK, HIF-1α and EPO proteins in C6 cells pretreated by hypoxia. The relationship between the tolerance against OGD induced by hypoxia preconditioning and the MEK/ERK pathway was elucidated in the pres- ence of PD98059 and LY294002. Results: Hypoxia preconditioning can induce protection against OGD. The most effective duration of hypoxia preconditioning against 24 h-OGD is 16 hours.
