凝聚胺抗体筛查技术的改良

     [摘要] 目的:对试管凝聚胺法进行进一步改良,建立一种简便、快速的无须离心的微孔板凝聚胺抗体筛查方法。方法:采用自制的5×10微孔板代替传统的试管,省却离心等步骤进行不规则抗体筛查。结果:将传统的试管凝聚胺法同无须离心的微孔板凝聚胺法进行比较,试管凝聚胺法检测抗D、抗E灵敏度与微孔板法无差异,抗C稍高于微孔板法(1∶512&1∶256);输血前筛查200份标本,同时发现2例不规则抗体,分别为抗D和抗M;对以前鉴定的11例不规则抗体,两者都漏检了抗Jka,其他完全一致。结论:无须离心的微孔板凝聚胺法无须试管及离心设备,具有简便快速、经济有效等特点,是一种可行的抗体筛查方法。

    Antibody Screening without a Centrifuge with Modified Polybrene Microplate Polybrene

    Abstract:Objective To establish a rapid and simple microplate Polybrene method without acentrifuge for antibody screening.Methods Modify the traditional tube Polybrene method,anddesign a special 5×10 microplate instead of traditional tube,without a centrifuge for antebodyscreening.Results Compared the tube Polybrene method with the microplate Polybrene method,the efficiency has no difference:antiD was 1 024 and antiE is 512,there were some difference about antiC,the former is 512,the later is 256.The antibody screening results were the same.Conclusion The microplate Polybrene method without a centrifuge was simple,rapid,economic and efficient method,what's more,which don't need tubes and centrifuging and it is feasible method for routine antibody screening. ......
