高效液相色谱法同时测定莲子心中四种异喹啉生物碱的含量

     摘要:目的:建立同时测定莲子心中四种异喹啉生物碱莲心季铵碱、莲心碱、异莲心碱、甲基莲心碱含量的离子对反相高效液相色谱(RPHPLC)法。方法:用十八烷基硅烷键合硅胶为填充剂,色谱柱:HibarTR,5 μm,250 mm×4.6 mm;流动相:乙腈水冰醋酸(55∶45∶1,每1 000 ml中加入4 g十二烷基磺酸钠);流速:1.2 ml/min;检测波长:282 nm;柱温:30℃。结果:莲心季铵碱在0.94~2.82 μg,莲心碱在1.14~3.42 μg,异莲心碱在0.7~2.1 μg,甲基莲心碱在0.96~2.88 μg范围内均呈良好线性关系;并均具有较好的加样回收率(>95%)。结论:该分析方法快速、准确、灵敏度高,重复性好,可用于同时对药材莲子心中四种异喹啉生物碱的含量测定。

    Determination of Four Isoquinoline Alkaloids in Plumula Nelumbini by HPLC Simultaneously

    Abstract:Objective:An ionpair RPHPLC method was established to determine the amounts of lotusine, liensinine, isoliensinine and neferine in Plumula Nelumbini(PNN)simultaneously.Methods:The column is packed with Hibar RT C18(5 μm, 250 mm×4.60 mm);The mobile phase consists of acetonitrile/H2O/glacial acetic acid(55∶45∶1),with 4 g sodium lauryl sulfonate in 1 liter, at the flow rate of 1.2 ml/min and the detection wavelength is 282 nm while the column temperature is 30℃.Results:Lotusine, liensinine, isoliensinine and neferine were proved to be linear within the range of 0.94~2.82 μg, 1.14~3.42 μg,0.7~2.1 μg and 0.96~2.88 μg respectively,with the recovery of the four alkaloids all being above 95%.Conclusion:This method is feasible to determine these four isoquinoline alkaloids simultaneously in PNN with the advantages of quickness, accuracy, high sensitivity and good repeatability. ......
