Using whole body clearance technique as well asisolated perfusedrat kidney (IPRKj. We studied theeffect of hyperglycemia on renal hemodynamics andthe possible mechanisminvolved. Hyperglycemia in-creased renal plasma flow (RPF)and glomerularfiltration rate (GFR). In nonfiltering isolated perfusedrat kidney or after adding furosemide to the perfusate,both techniques were believed to abolish thetubuloglomerular feedback, the renal hemodynamicchanges induced by hyperglyeemia disappeared, butadding sodium nitroprusside increased RPF and de-creased renal vascular resistanee (RVR). D-a-methyl-glucoside, a glucose derivative, increased RPFand GFR in IPRK, but had np influence onrenalhemodynamics in whole body clearance study.Hemoglobin, which could block the action ofendotheliumderived relaxing factor (EDRF), did notchange the effects of hyperglycemia on renalhemodynamics in IPRK.These results indicated thathyperglyeemia could directly induce hyperl)erfpsionand hyperfiltration, which is mainly mediated by sup-pression of tubuloglomerular feedback. EDRF do notplay the major role in the changes of hemodynamicsinduced by hyperglycemia.

Protective effect of 1,6-dipbospkate fructose in boratoryischeinic renal failure in elderly rats Li Liying, WangHaiyan, Shi fianhua, et al. Neforotogy institute, Beijing Medical University,Beilng 100034
To study the protective effect of l, 6-diphosphatefructose (FDP) on acute ischemia damage in brain,heart and liver, we observed its effect on acuteischemic renal failure fAIRF). AIRF in male Wisterrats was produced by renal artenal clamping for 45minutes and reperfusion for 90 minutes. The rats weredivided into 4 groups according to their age and FDPtherapy: GI : young (3 - 4 months) AIRF rats withoutFDP; G2. old (26-27 months) AIRF rats withoutFDP; G3. young AIRF rats with FDP (o.sg / kg) ivinfusion, G4 old AIRF rats with FDP. Inulin clearance (Cin), PAH clearance (Cpah), urinary sodium(UNA) were examined before ischemia and 30, 60 and90 minutes after reperfusion. After reperfusion for 90minutes, renal cortex was taken for the examination ofrenal mitochondrial ATP synthetic content (miATPs).Ci.and Cpah were lower in G2 than in GI (0.24 ± 0. 1 2vs l.75± 0.79, P<0.0l, 0.87±0.42 vs 7.12± 4.04,P<0.05, at 90 minutes reperfusion). Cin and Cpah inGI showed an autorecovery up to 40.1 % and 33.7%of the level in contrast to G2. In G3 and G4 originalFDP infusion resulted in a significant increase inmiATPs and C,.(609 ± 145 vs 389 ± 97, 569 ± 77 vs 338± 61, P< 0.02, 2.47 ± 0.58 vs I .75 ± 0.79, l .29 ± 0.42 vs0.24± 0.12, P<0.0l). CIn of the old was higher thanthat of the young and the miATPs increased more evi-dently in the old than in the young (6.0 vs 3.5 times.and 68.% vs 57%). These data suggest that FDPbrings about greater protective effect on the old thanon the young from AIRF, which might relate to itsrole of energe providing under anaerobic glycolysiscondition.

To observe the changes of oxygen dynamic statusin the development of chronic cor pulmonale, We stu-died early chronic cor pulmonale fgroup A, 13 cases),late chronic cor pulmonale in remission stage tgroupB, 15) and late chronic cor pulmonale in acuteexcerbation stage (group C, 10) with SwanGanzthermodilution catheter and arterial and mixed venousblood gas analysis for detecting the oxygen deliveryfDO,) and consumption (VO2). In group C. amrinonewas given intravenously (lspg . kg i / min) and theexaminations were repeated. DOz (465± 72ml .mini / mz) and VO, (l14+ I lml . mini / mz) wereat low level of normal range. No correlation was notedbetween them (r=0.282, P>0.05) in group A Itissuggested that the patient of early stage was in low ox-ygen delivery and consumption and no tissue oxygendeficit existed. In group B, DOz andVOz increasedf592±111 and 145 ± 32ml . min1 / mz respectively.P < 0.05 versus group A) but no significant correlationwas found (r = 0. 1 89, P > 0.05). Hence, the body of thepatient was in high metabolic status and no tissue ox-ygen decifit existed in remission stage. DOz andVO, in group C(622± 150, 139± 44ml.mini/mz respectively) were similar to those in groupBfP<0.05). After amnnone administration, a signifi-cant correlation was found between DO2 andVO, (r＝0.39, P<0.05). It is suggested that in theexcerbation stage, the patient's body was in highlymetabolic status, the oxygen deliveryconsumptioncompensation mechanism failed, and tissue oxygendeficit existed.

In 280 patients with IDDM, the positive familyhistorV of diabetes was 26.8% of IDDM probands.The prevalence of diabetes in relatives was 68 % in firstdegree relatives, 28% in second degree relatives, and4% in third degree relatives. HLA typing of 87 mem-bers in 13 pedigrees with IDDM was performed. Thesedata support the hypothesis that IDDM is amultigenic hereditary disorder.

In 87 patients with peripheral pulmonary diseaseseither adhesive or close to the pleura. who were exam-ined, ultrasonographically 64 had lung cancer and 23benign lesions were confirmed histopathologically.Five sonographic features including configuration.echogenicity patterns, bronchial air phase in foci,margin, and pleural involvement were significantly different between malignant and benign diseases(P<0.ol). The differential ultrasonography used inthis series showed a sensitivity of 61% (14/ 23), aspecificity of 95% (61 / 64), and an accuracy of 86%(75 / 87). The results demonstrate that ultrasound ishelpful in differentiating the malignant lesion from thebenign one in the peripheral lung near the pleura.

To clarify the relationship between theneuroendocnne expression of prostatic carcinoma andprognosis, we studied immunohistochemicallyformalinfixed and paraffinembeded tissue sectionsfrom 30 cases of prostate cancer by means ofneuroendocrine (NE) differentiation markers. The rateof N E positivestaining cases was 40% (l2/30), andmore than two kinds of NE markers were expressed in9 / 12 cases. The NE markers were NSE (7 positivecases), ChromograninA (7), Calcitonin fZ), GastrinfZ), hCG (4), S100 Protein (2), Serotonin (3j, EMA(23) and Lillie argentaffin staining fZ). Followup ofthe 30 patients showed that NE negative tumors havea prognosis better than NE positive tumors(P<0.005). Their poor prognosis is due to poor re-sponse to endocrine therapy, and the mechanism isthat some neuropeptides secrected by NE cells affectthe growth of tumor cells. Finally, how to improve theprognosis of the patients who have NE expression oftheir prostate cancer is discussed.

Circulatory crisis occurred in 2 cases oftoetohand transfer and I case of musculocutaneousfree flap after smoking. Two of them failed and I sur-vived after exceeding antispasm and antithrombosistreatment. Experimental studies showed hannful in-fluences of cigarette smoking on endothelial healingprocess at the vascular anastomosis site, which wascovered bv endothelial eells in 16%-32% andplateletin 8%-10% of the smoking group 5-10 daysafter operation, but in 75%-100% and 0% of thenornial group. The mechanism and characteristics ofcirculatory crisis caused by cigarette smoking are dis-cussed.

The concentration of urinary nicotine ofnonsmoking female workers. 56 from shreddingworkshop, 65 from packing workshop and 54 frombeverage factory (as controls), was determined, and intervention test for the absorption of nicotine was car-ried out in female workers from shredding workshop.The mean geometric concentration of urinary nicotineof shredding workers and packing workers at the endof shift was 44.8 ± 5. 1ng /ml and 9.9 ± 5.4ng / ml, andwas significantly higher than that of the controls (l .0± 1 .ong / ml) (P < 0.01). The concentration of urinarynicotine of shredding workers was significantly higherat the end of shift than preexposure period (11.46±6.73ng/ml). The intervention test for the absorptionof nicotine showed that there were good protection effects for the absorption of nicotine when glove andmask were used simultaneously and ventilator wasused in summer.

The loss of heterozygosity (LOH) at chromosomes 3p24, 7q31.16q24.3and17p13.1was measuredin 15 patients with breast cancer by means ofpolymerase chain reaction (PCR)-restriction fragmentlength polymorphisms fRFLPj or PCRCA repeatanalysis. In 36%, 36%, 50% and 56% of the informa-tive cases, LOH was detected at 3p24, 7q31, 16q24.3and 17pl3. 1 loci, respectively. All the LOH at 3p24 or7q31 happened in stage II or III, 67% of the LOH at3p24 or 7q31 happened in the cases withlymph node metastasis, and 67% of the LOH at 3p24or 7q31 were found in estrogen receptor negative pa-tients. This finding shows that the measurement ofLOH at 3p24 and 7q31 may be an useful predictingmarker for the prognosis of patients with breastcancer. The high percent of LOH at these four locimay also indicate the possibility of the existence oftumor suppressor genes for breast cancer.

Hereditary glucose-6-phosphate dehydrogenase(G6PD) in red blood cell was one of the most commongenetic diseases in South China. The research data ofG6PD gene showed that at least 6 point mutationswere responsible for various G6PD variants inChinese. For developing a rapid, sensitive, and effec-tive method to detect point mutation, we applied sin-glestrand confoimation (SSCP) analysis for detectionof mutation in exon 2 of G6PD gene of 20 cases ofG6PD variants. Four of them were found that mobili-ty shift band in one of two single strands DNA isslower than other individuals. PCR direct sequencingfor these 4 samples were given a base sustitution (T toC) at nucleotide 95 of cDNA. The results indicatedthat this technique is very simple, sensitive, and usefulover other methods of detecting point mutation. Theexperimental conditions of PCRSSCP and featuresof this mutation were discussed at the same time.

Endotoxin was as a pathogen to investigate its ef.fects on bronchial hyperreactivity formation. The la-tent peroids of guinea pigs inhaled histamine andPDz value of isoproterenol were markedly decreased 4days after the intraperitoneal injection of endotoxin Img/kg, meanwhile PD2 value of histamine andhistamine levels of lung tissue and plasma were in-creased significantly. These results indicate thatendotoxin is a very important factor in the infectioninduced bronchial hyperreactivity.

The intervertebral distance in 31 prolapsed discsunder traction increased on the average by l.34mm.but in nonnal discs 0.87mm. Of 47 prolapsed discswithout traction, 29 had negative intradis pressure, 10zero, and 8 positive. The total average value of thepressure before traction was - 1.3 kpa. Under lumbertraction, the pressure decreased in 30 discs, unchangedin 8, and increased in 9, with the average value being3.2 kpa.The intradisc pressure in 7 normal discs wascontrary to the pressure in the prolapsed disc. The dif-ference between the two groups was highly significant(P< 0.01). The pressure unchanged when theintervertebral distance of prolapsed disc unchangedunder traction, but the pressure decreased as the dis-tance increased in most cases under traction.

The concentration of urinary nicotine ofnonsmoking female workers. 56 from shreddingworkshop, 65 from packing workshop and 54 frombeverage factory (as controls), was determined, and intervention test for the absorption of nicotine was car-ried out in female workers from shredding workshop.The mean geometric concentration of urinary nicotineof shredding workers and packing workers at the endof shift was 44.8 ± 5. 1ng /ml and 9.9 ± 5.4ng / ml, andwas significantly higher than that of the controls (l .0± 1 .ong / ml) (P < 0.01). The concentration of urinarynicotine of shredding workers was significantly higherat the end of shift than preexposure period (11.46±6.73ng/ml). The intervention test for the absorptionof nicotine showed that there were good protection effects for the absorption of nicotine when glove andmask were used simultaneously and ventilator wasused in summer.

The loss of heterozygosity (LOH) at chromosomes 3p24, 7q31.16q24.3and17p13.1was measuredin 15 patients with breast cancer by means ofpolymerase chain reaction (PCR)-restriction fragmentlength polymorphisms fRFLPj or PCRCA repeatanalysis. In 36%, 36%, 50% and 56% of the informa-tive cases, LOH was detected at 3p24, 7q31, 16q24.3and 17pl3. 1 loci, respectively. All the LOH at 3p24 or7q31 happened in stage II or III, 67% of the LOH at3p24 or 7q31 happened in the cases withlymph node metastasis, and 67% of the LOH at 3p24or 7q31 were found in estrogen receptor negative pa-tients. This finding shows that the measurement ofLOH at 3p24 and 7q31 may be an useful predictingmarker for the prognosis of patients with breastcancer. The high percent of LOH at these four locimay also indicate the possibility of the existence oftumor suppressor genes for breast cancer.

Hereditary glucose-6-phosphate dehydrogenase(G6PD) in red blood cell was one of the most commongenetic diseases in South China. The research data ofG6PD gene showed that at least 6 point mutationswere responsible for various G6PD variants inChinese. For developing a rapid, sensitive, and effec-tive method to detect point mutation, we applied sin-glestrand confoimation (SSCP) analysis for detectionof mutation in exon 2 of G6PD gene of 20 cases ofG6PD variants. Four of them were found that mobili-ty shift band in one of two single strands DNA isslower than other individuals. PCR direct sequencingfor these 4 samples were given a base sustitution (T toC) at nucleotide 95 of cDNA. The results indicatedthat this technique is very simple, sensitive, and usefulover other methods of detecting point mutation. Theexperimental conditions of PCRSSCP and featuresof this mutation were discussed at the same time.

Endotoxin was as a pathogen to investigate its ef.fects on bronchial hyperreactivity formation. The la-tent peroids of guinea pigs inhaled histamine andPDz value of isoproterenol were markedly decreased 4days after the intraperitoneal injection of endotoxin Img/kg, meanwhile PD2 value of histamine andhistamine levels of lung tissue and plasma were in-creased significantly. These results indicate thatendotoxin is a very important factor in the infectioninduced bronchial hyperreactivity.

The intervertebral distance in 31 prolapsed discsunder traction increased on the average by l.34mm.but in nonnal discs 0.87mm. Of 47 prolapsed discswithout traction, 29 had negative intradis pressure, 10zero, and 8 positive. The total average value of thepressure before traction was - 1.3 kpa. Under lumbertraction, the pressure decreased in 30 discs, unchangedin 8, and increased in 9, with the average value being3.2 kpa.The intradisc pressure in 7 normal discs wascontrary to the pressure in the prolapsed disc. The dif-ference between the two groups was highly significant(P< 0.01). The pressure unchanged when theintervertebral distance of prolapsed disc unchangedunder traction, but the pressure decreased as the dis-tance increased in most cases under traction.
