多奈哌齐与维拉帕米联合用药对大鼠阿尔茨海默病模型血液流变学的影响(1)

[摘要]目的观察多奈哌齐与维拉帕米联合用药对大鼠阿尔茨海默病模型(AD)血液流变学的影响。方法清洁级健康成年雄性SD大鼠40只，(250±30)g，随机均分至对照组、模型组、多奈哌齐组、多奈哌齐+维拉帕米组(以下简称为联合用药组)。用脑立体定位仪将致脑内神经元胞外淀粉变样的Aβ1-40 (2 μg/μl)5 μl注入右侧海马造成AD模型；对照组则注射等量注射生理盐水。正常喂养7 d后，灌胃给药，多奈哌齐组用多奈哌齐(6.79×10-2mg/ml)灌胃，联合用药组用多奈哌齐(6.79×10-2mg/ml) 和维拉帕米(3.825×10-1mg/ml)灌胃，对照组和模型组均用等量双蒸水灌胃，1次/d，持续28 d。即后处死，腹主动脉采血，分别测定血液流变学的各项指标。结果模型组大鼠的多项血液流变学指标均显著高于对照组、多奈哌齐组及联合用药组(P<0.05)；联合用药组在降低AD大鼠全血粘度(高切、低切)、红细胞刚性指数方面与多奈哌齐组比较具有统计学意义，在降低红细胞压积、聚集指数方面优于多奈哌齐组；对照组、多奈哌齐组及联合用药组的多项血液流变学指标差异不具有显著性。结论AD大鼠存在血液流变学异常，多奈哌齐与维拉帕米联合用药能改善AD大鼠的血液流变学指标且优于单纯应用多奈哌齐。

[Abstract]ObjectiveTo observe the effect of Donepezil combined Verapamil on Hemorheology of rat of Alzheimer's disease. MethodsForty cleaning healthy grown-up male rats,250±30g,were randomly divided equally into the control group、model group、Donepezil group、Donepezil+ Verapamil(drug combination group) .Aβ1-40 (2μg/μl)5μl was infused in the hippocamp of right for AD model by stereotaxis instrument;the equivalent NS for injection was infused in the same site for control group.All of the rats had been feeding normally for seven days then administration by gavage. The rats of Donepezil group were lavagedDonepezil(6.79×10-2mg/ml);the rats of drug combination group were lavaged Donepezil(6.79×10-2mg/ml)and Verapamil(3.825×10-1mg/ml);the rats of control and model groups were lavaged equivalent double distilled water,one time a day for twenty eight days.Then they were executed and sampled blood through abdominal aortas and determined the indexes ofhemorheology respectively.Results Model group's most indexes of hemorheology showed significantly higher than those of control group、Donepezil group and drug combination group(P<0 ......
