麻杏石甘汤对哮喘大鼠气道上皮细胞粘蛋白表达的影响研究(1)

     摘要:目的 研究麻杏石甘汤对哮喘大鼠气道上皮细胞粘蛋白表达的影响,探讨其在哮喘粘液高分泌中的作用。方法 将44只SPF级雄性SD大鼠随机分为四组,对照组(A组),哮喘组(B组),哮喘+布地奈德组(C组),哮喘+麻杏石甘汤组(D组)。留取支气管肺泡灌洗液(BALF)检测其中IL-12、IL-13的浓度;切取肺组织标本行HE染色观察气道炎症,AB-PAS法检测各组大鼠气道粘蛋白的表达,检测吸光度值后运用SPSS17.0进行统计。结果 ①麻杏石甘汤能降低哮喘组BALF中IL-13浓度,上调IL-12含量;②麻杏石甘汤能减轻哮喘大鼠气道及血管肌层增生情况,减少周围炎性细胞浸;③哮喘组存在气道粘液高分泌,麻杏石甘汤能下调气道粘蛋白的表达,但作用较布地奈德组弱。结论 麻杏石甘汤可减少粘蛋白表达,减轻哮喘大鼠炎症反应、气道粘液高分泌,为其在哮喘的现代临床应用提供依据。

    Effect of Maxingshigang Decoction on Expression of Mucin in Airway Epithelial Cells of Asthmatic Rats

    Abstract:Objective To study the effect of Maxingshigan decoction on expression of airway epithelial cell mucin in asthmatic rats and explore its role in mucus hypersecretion in asthma.Methods 44 male SD SPF rats were randomly divided into 4 groups,the control group(A group),asthma group(B group),Asthma+budesonide group(C group),asthma+apricot stone group(D group).Take the bronchoalveolar lavage fluid(BALF)to detect the IL-12,IL-13 concentration;cut the lung tissue HE staining observation of airway inflammation,examined the expression of rat airway mucin AB-PAS method, detecting the absorbance after using SPSS17.0 statistics.Results ① Maxingshigan decoction could decrease the concentration of IL-13 in BALF of asthmatic group and up-regulate the content of IL-12.② Maxingshigan decoction can reduce the airway and vascular myometrial hyperplasia of asthmatic rats,reduce the surrounding inflammatory cell leaching.③ Asthmatic group had a high secretion of airway mucus,and Maxingshigan decoction could down-regulate the expression of airway mucin,but the effect was weaker than that of budesonide group.Conclusion Maxingshigan decoction can reduce the expression of mucin,reduce the inflammatory reaction and airway mucus secretion in asthmatic rats,and provide the basis for its clinical application in asthma.

    支气管哮喘(简称哮喘,bronchial asthma)是儿童时期最常见的慢性疾病之一[1],发病率逐年攀升,其发病与气道慢性炎症、气道高反应(airway hyperresponsiveness,AHR)、气道重塑、粘液高分泌等密切相关。过度的气道粘液分泌提示哮喘控制不佳,是许多哮喘患兒的共同临床特点,已经成为影响哮喘病情和预后的独立危险因素,但在哮喘发病中的作用往往被忽视[2]。减少粘液分泌、促进粘液排出是哮喘防治亟待解决的重要课题。麻杏石甘汤出自经典国方《伤寒论》,由麻黄、杏仁、炙甘草、石膏四味药组成,是治疗热哮的良方。现代药理学研究表明其具有解热平喘、镇咳祛痰、抗炎、抗过敏的功效[3]。但其在哮喘防治中的具体机制仍未阐明,对哮喘气道粘液分泌的影响更是鲜有报道。因此,本研究拟通过建立哮喘大鼠模型,采用AB-PAS染色法等研究气道粘蛋白表达,探讨麻杏石甘汤在哮喘粘液高分泌中的作用。, http://www.100md.com(虞琳 潘建伟)
