补肾复方对压力负荷增加大鼠心肌纤维化的影响(1)

摘 要:目的:观察补肾复方逆转压力负荷增加大鼠心肌纤维化的作用。方法:采用腹主动脉狭窄所致压力负荷增加大鼠左室重构模型,将雄性SD大鼠60只随机分为假手术组、模型组、卡托普利组、补肾复方大剂量组、补肾复方小剂量组,观察用药4周后左室心肌病理形态胶原染色、左室心肌间质超微结构、左室心肌Ⅰ型胶原免疫组织化学染色、左室心肌Ⅲ型胶原免疫组织化学染色等指标的改变。结果:模型组Ⅰ型胶原的平均光密度与假手术组比显著升高(P<0.01),卡托普利组、补肾复方大剂量组、小剂量组的Ⅰ型胶原平均光密度均较模型组显著下降(P<0.01)。模型组Ⅲ型胶原的平均光密度与假手术组比显著升高(P<0.01),卡托普利组、补肾复方大剂量组、小剂量组的Ⅲ型胶原平均光密度均较模型组显著下降(P<0.01)。左室心肌病理形态胶原染色、左室心肌间质超微结构的改变与Ⅰ型胶原和Ⅲ型胶原的改变基本一致。结论:补肾复方具有与卡托普利基本一致的逆转心肌纤维化的作用。

Abstract:Objective:To observe the effects of Bu Shen Prescription on regressing myocardial fibrosis in rat with pressure overload. Methods:The model of left ventricular remodeling in rat with pressure overload by abdominal aortic constriction was used. 60 SD male rats were randomly divided into five groups: sham operation group, model group, captopril group, Bu Shen Prescription high dosage group and low dosage group. After treatment 4 weeks, it was observed that the changes of left ventricular myocardium collagen pathological coloring, left ventricular interstitial cardiac tissue ultramicrostructure, left ventricular cardiac tissue collagen type Ⅰimmunohistochemistry coloring and collagen type Ⅲ immunohistochemistry coloring. Results: The average optic density of left ventricular cardiac tissue type Ⅰcollagen and type Ⅲ collagen of model group were increased obviously than that of sham operation group (P<0.01). The average optic density of left ventricular cardiac tissue type Ⅰcollagen and type Ⅲ collagen of captopril group, Bu Shen Prescription high dosage group and low dosage group were lower than those of model group significantly(P<0.01). The changes of left ventricular myocardium collagen pathological coloring and left ventricular interstitial cardiac tissue ultramicrostructure were consistent with those of typeⅠ collagen and type Ⅲ collagen ......
