丹参联合骨髓间充质干细胞静脉移植对兔心肌梗死区血管再生影响的实验研究(1)

摘 要:目的:探讨骨髓间充质干细胞静脉移植对兔急性心肌梗死(AMI)梗死区及梗死周围区血管新生的影响及中药丹参注射液对其的干预作用。方法:用开胸结扎冠状动脉左室支的方法建立急性心肌梗死模型,将已建立心肌梗死模型的日本大耳白兔随机分为3组,模型对照组、静脉移植骨髓间充质干细胞组、静脉移植骨髓间充值干细胞联合丹参组。术后5周取材,观察梗死区内CD31(血小板/内皮细胞黏附分子)和VEGF(血管内皮生长因子)蛋白的表达。结果:1免疫组化CD31表达,静脉注射干细胞组和干细胞联合静脉注射丹参组高于模型组(P<0.05);2免疫组化VEGF蛋白表达,各组间无统计学差异(P>0.05)。3血清中VEGF蛋白含量,各组间无统计学差异(P>0.05)。结论:静脉注射骨髓间充质干细胞可诱导梗死区血管新生,其机制并非通过VEGF的表达来调节的,可能是通过其他途径调节的,中药丹参注射液对干细胞促进血管新生无明显促进作用。

Abstract:Objective: To investigate bone marrow stem cells in vein graft result in rabbits with acute myocardial infarction (AMI) in peri-infarct area and infarct angiogenesis effect of Chinese medicine Danshen injection on the role of its intervention. Methods: The left ventricular branch coronary artery ligation thoracotomy approach model of acute myocardial infarction, myocardial infarction model will have established a Japanese white rabbits were randomly divided into three groups, model control group, intravenous transplantation of bone marrow mesenchymal stem cell group, Danshen injection combined with intravenous transplantation of bone marrow-derived mesenchymal stem cell group. Drawn after 5 weeks,Infarct area observed CD31 (platelet / endothelial cell adhesion molecule) and VEGF (Vascular Endothelial Growth Factor) protein expression.
