亚麻木酚素对糖尿病小鼠肾脏保护作用及机制(1)

    摘 要:目的:观察亚麻木酚素提取物(FLE)对糖尿病小鼠早期肾损伤的影响并探讨其作用机制。方法:雄性ICR小鼠分6组:正常对照组,糖尿病模型对照组,糖尿病模型亚麻木酚素提取物[600mg/(kg•d)、1.2g/(kg•d)、3.6g/(kg•d)]治疗组,糖尿病小鼠盐酸二甲双胍[100mg/( kg•d)]治疗组。连续灌胃8周,实验结束后比较各组体重、肾重、肾重/体重、24h尿白蛋白排泄率、肾组织MDA含量及肾组织SOD、GSH-PX活性,HE染色分析肾小球病理形态学改变,综合评价亚麻木酚素提取物对肾脏的保护作用。结果:与糖尿病模型对照组比较,亚麻木酚素提取物中、高剂量组的肾重、24h尿白蛋白排泄率显著降低(P<0.05),MDA含量降低(P<0.05),亚麻木酚素提取物各剂量组的肾脏组织SOD、GSH-Px活性增加(P<0.05)。结论:亚麻木酚素提取物能减轻糖尿病小鼠肾脏损伤,对肾脏有保护作用,其机制与其减轻肾脏氧化应激有关。

    Study on the Protective Effect of Flaxseed Lignan Extract on Kidney in Diabetic Mice

    Abstract:Objective: To investigate the protective effect of of Flaxseed Lignan Extract(FLE)on kidney in diaetic mice. Methods: ICR mice were randomly divided into 6 groups: normal mice control, STZ diabetic mice control, STZ diabetic mice fed with FLE at dose of [600mg/(kg•d),1.2g/(kg•d), 3.6g/(kg•d)], and STZ diabetic mice fed with metformin. The above components were administed intragastrically for 8 weeks. Then their blood glucose, kidney weight to body weight, 24-hour urinary protein,the content of MDA and the activities of SOD, GSH-Px of kidney were detected. Glomerular structure of kidney were examined by light microscopy. Results:The kidney weight to body weight, 24-hour urinary protein in high,middle dose of FLE groups were significantly lower than those in diabetic control group(P<0.05).
