葛花水提取液对蚕豆根尖细胞微核率的影响

     摘要:目的探讨葛花水提取液对蚕豆根尖细胞微核率的影响。方法用不同浓度的葛花水提取液处理未添加或添加环磷酰胺溶液的蚕豆根尖样品,经镜检测算细胞微核率。结果葛花水提取液浓度为1.0 g﹒mL-1时,由自然诱导或环磷酰胺诱导的蚕豆根尖细胞微核率最低,分别为4.40 ‰和11.11 ‰;对蚕豆根尖细胞微核的抑制率达到最高,分别为79.07%和53.90%。结论葛花水提取液可有效地抑制蚕豆根尖细胞微核的产生,且抑制作用随着葛花水提取液浓度的增大而增强,表明葛花水提取液具有抑制遗传毒性的作用。

    Effect of Water Extract of Flos Puerariae on Micronucleus Formation in Vicia faba Root Tip Cells

    Abstract:Objective To approach the effect of water extract of Flos Puerariae on micronucleus formation in Vicia faba root tip cells. Methods The root tips of Vicia faba with cyclophosphamide or without cyclophosphamide were treated with different concentrations of water extract of Flos Puerariae and the microscope was used for the examination of micronucleus formation. Results When the concentration of water extract of Flos Puerariae was 1.0 g﹒mL-1, the micronucleus formation(MCN‰)in root tip cells which was created naturally or induced with cyclophosphamide was lowest, 4.40‰ and 11.11‰ respectively; the inhibition rate on micronucleus formation was tallest, 79.07% and 53.90%, respectively. Conclusion The water extract of Flos Puerariae can effectively inhibit the creation of root tip cells micronucleus of Vicia faba and the inhibitory action of the water extract of Flos Puerariae can be strengthened along with the increasing concentrations of itself, which indicates the water extract of Flos Puerariae has an inhibitory effect on hereditary toxicity.

    葛花(Flos Puerariae)为豆科植物野葛[Pueraria lobata (Willd.) Ohwi]、甘葛藤(Pueraria thomsonii Benth.)的花,异名葛条花[1],具有解酒醒脾的功能,能有效抑制伤酒发热烦渴、不思饮食、呕逆吐酸、吐血、肠风下血等症状[2]。现代药理研究表明,葛花可降低血液中的乙醇浓度[3]。国内学者曾开发了以葛花为主要材料的醒酒保健品“葛花露”。葛花还具有保护肝脏以及调节血糖、血脂的作用,兼具抗肿瘤、抗诱变、抗幽门螺杆菌、抗过敏等功效。

    目前对葛花的研究多为其解酒功能及其黄酮、葛花苷等化学成分,对其水提取液遗传毒性的研究很少。我们通过蚕豆根尖微核实验,研究了葛花水提取液对环磷酰胺诱导及自然诱导的蚕豆根尖微核率的影响,揭示了葛花水提取液抑制遗传毒性方面的功能,此研究结果将使此传统药物在防病治病中起更大的作用。
