川芎提取液对小白鼠离体子宫平滑肌收缩性能的影响

     [摘要] 目的:观察不同浓度的川芎提取液对小白鼠离体子宫平滑肌收缩性能的影响。方法:30只小白鼠随机分为3组:高浓度组、中浓度组和低浓度组,每组10只;实验前3 d,每天皮下注射己烯雌酚0.2 ml/只。实验时颈椎脱臼处死小白鼠,剖宫取出子宫,置于与压力换能器相连的麦氏浴槽中,持续供氧,记录正常收缩曲线10 min,然后加入不同浓度的川芎提取液,分别记录加入川芎提取液后子宫的活动曲线。结果:川芎提取液对小白鼠离体子宫平滑肌具有抑制作用,浓度越大,抑制作用越明显(P<0.05)。结论:川芎提取液对子宫平滑肌具有明显的抑制作用。

    Influence of Chuanxiong Extract on the contraction performance of in vitro mice uterus smooth muscle

    [Abstract] Objective: To observe Chuanxiong Extract on the contraction performance of in vitro mice uterus smooth muscle. Methods: 30 mice were randomly divided into 3 groups: high concentration group, moderate concentration group and low concentration group, with 10 mice in each group. 3 days before the experiment, 0.2 ml Stilbestrol were given to each group every day via subcutaneous injection. Then the mice were killed through cervical dislocation, and the uterus was taken out and placed in culture medium, then put into pressure transducer connected with Michael's bath, continued supplying oxygen. The normal curve of the uterine contraction was recorded for 10 min, then different concentrations of Chuanxiong Extract were added, the activity curves were recorded separately after adding Chuanxiong Extract. Results: Chuanxiong Extract had an inhibitory effect on in vitro uterine smooth muscle; with the increase of concentration, the inhibitory effect was more obviously (P<0.05). Conclusion: Chuanxiong Extract has a significantly inhibitory effect on uterine smooth muscle.

    川芎为伞形多年生草本植物,性辛,温,无毒,入肝、胆、心包经,含挥发油和内酯化合物如川芎内酯、川芎酚,生物碱如川芎嗪等,其生物总碱及川芎嗪具有显著的强心作用,使心肌收缩力加强,心率加快,心输出量增加,周围血管扩张,血压下降,心脑血管血流量增加,微循环改善,对心肌缺血-再灌注损伤有明显的保护作用[1],但川芎提取液用于实验性拮抗小白鼠离体子宫收缩作用的研究很少。本研究旨在通过测定小白鼠离体子宫加入川芎提取液后子宫活动曲线的变化,为临床上更好地利用川芎提供参考资料。

    1 材料与方法

    1.1 实验动物

    雌性小白鼠30只,体重18~22 g,由右江民族医学院实验动物中心提供。随机分为3组,每组10只,分为高浓度组、中浓度组和低浓度组。

    1.2 实验材料与药品

    1.2.1 仪器与器械BL-410生物功能实验系统、张力换能器、超级恒温水浴器(上海智城分析仪器制造有限公司)、麦氏浴槽、通气管、小弯钩、氧气球胆、小镊子、小剪刀、组织剪、培养皿、1 ml注射器。

    1.2.2 药品乐氏液(NaCl 9.2 g,10% KCl 4.2 ml,10% CaCl2 2.4 ml,5% NaHCO3 3.0 ml,葡萄糖1.0 g,加蒸馏水至1 000 ml),0.2%己烯雌酚,川芎提取液(自备):川芎干品100 g,用自来水浸泡4 h,加自来水至1 000 ml煮沸1 h,过滤,滤渣加水至1 000 ml,煮沸1 h,过滤,弃滤渣,合并2次滤液,过滤,加入95%乙醇溶液100 ml,醇提,过滤得100 ml,含生药1 g/ml。

    1.3 装置

    将麦氏浴槽与BL-410生物功能实验系统相连接,放入恒温浴槽中,温度为(36.0±0.5)℃,实验时持续供氧。

    1.4 实验步骤

    小白鼠离体子宫实验[2]:将小白鼠分笼饲养,自由饮食,室温(28±2)℃。在实验前3 d,每天皮下注射己烯雌酚0.2 ml/只,注射3 d后将小白鼠颈椎脱臼,剖宫取出子宫,立即放入乐氏液中,轻剥子宫壁上的脂肪和结缔组织,从子宫角剪开,用线结扎两端,子宫的一端固定在通气钩上,另一端和张力换能器相连接,然后将通气管放入盛有营养液的麦氏浴槽中,使营养液的体积固定在50 ml,张力换能器负荷为1.5 g。用BL-410生物功能实验系统描记子宫活动曲线 ......
