La(C7H5O3)2.(C9H6NO)抑制Hela细胞生长作用机制研究(1)

     [摘要] 目的:观察La(C7H5O3)2·(C9H6NO)对体外培养的Hela细胞的生长抑制作用,并进一步探讨其作用机制。方法:应用MTT法检测细胞增殖活性,流式细胞仪检测细胞周期分布,AnnexinⅤ-FITC染色法检测细胞凋亡率。结果:不同浓度的La(C7H5O3)2·(C9H6NO)处理Hela细胞24~96 h后,细胞增殖受到显著抑制,并呈浓度及时间依赖性;0.5、1.0、5.0、10.0 μmol/L La(C7H5O3)2·(C9H6NO)处理72 h后,Hela细胞G0/G1期细胞数量显著增加,S期细胞数量显著减少(P<0.01);各浓度组细胞凋亡率均显著增加(P<0.01)。结论:La(C7H5O3)2·(C9H6NO)对Hela细胞具有生长抑制作用,其机制与阻滞细胞周期及诱导凋亡有关。

    Study on the inhibitory mechanism of La(C7H5O3)2·(C9H6NO) in the proliferation of Hela cells

    [Abstract] Objective: To investigate the inhibitory effect of La(C7H5O3)2·(C9H6NO) in the proliferation of Hela cells, and explore its mechanism. Methods: MTT assay, flow cytometry and AnnexinⅤ-FITC staining were respectively performed to determine the cell proliferation activity, cell cycle and apoptotic rate. Results: After treated with different concentrations of La(C7H5O3)2·(C9H6NO) for 24 to 96 hours, the proliferation activity of Hela cells was inhibited in a dose- and time-dependent manner. Hela cells were treated with 0.5, 1.0, 5.0, 10.0 μmol/L of La(C7H5O3)2·(C9H6NO), the number of cells in G0/G1 phase significantly increased and the number of cells in S phase significantly increased in cells treated with different concentrations of La(C7H5O3)2·(C9H6NO) (P<0.01). Conclusion: La(C7H5O3)2·(C9H6NO) can inhibit the growth of Hela cells by blocking cell cycle and inducing apoptosis.
