爬山虎籽总黄酮提取及鉴别(1)

     [摘要] 目的:研究爬山虎中黄酮的鉴别及总黄酮的提取工艺。方法:采用传统溶剂浸提法进行常规的化学定性分析确定爬山虎籽中含有黄酮类化合物,并采用五因素四水平的正交试验优化提取工艺。结果:实验证实爬山虎中含有大量黄酮类化合物。正交实验得出了最佳提取工艺条件:提取温度为50℃,料液比为1∶12,提取时间为3 h,提取剂为pH=2的65%乙醇溶液。以芦丁为标准品,用紫外分光光度法测定爬山虎籽中总黄酮含量为2.3%。结论:爬山虎籽中含有丰富的黄酮类化合物,优化并建立了爬山虎籽中总黄酮的提取工艺条件。

    Extraction techniques and identification of flavonoids in parthenocissus seeds

    [Abstract] Objective: To make a comprehensive study on identification and extraction technology of flavonoids from the parthenocissus seeds. Methods: Traditional solvent extraction method and orthogonal experiments were used. Flavonoids in the parthenocissus seeds were confirmed by conventional qualitative analysis. Results: The research showed that the parthenocissus seeds contain flavonoids. The optimum technical condition were summed up as follows through the orthogonal experiments: extraction temperature was at 50℃, proportion material and extracting reagent was 1∶12, the extracting time was 3 h, extracting reagent was 65% aqueous ethanol solution, and pH=2. Comparing to ruitin standard, extraction rate of flavonids in parthenocissus seeds was 2.3% by UV spectrophotometry. Conclusion: Flavonoids is rich in the parthenocissus seeds. The preliminary extracting technology of flavonoids from the parthenocissus seeds is established in the study.

    黄酮类化合物是天然药用植物的主要活性成分之一,其众多的生理功能使其在医疗方面应用较为广泛,可以预防高血压,具有保肝、降血脂、降胆固醇等多种功能[1-3]。此外,它还是重要的功能食品添加剂、天然抗氧化剂、天然色素,对它的开发研究已引起了国内外的广泛重视,研究进展很快[3]。爬山虎(Parthenocissus tricuspidate plach)属于葡萄科爬山虎属落叶多年生木质藤本攀缘植物[4]。爬山虎的根茎可入药,果实可酿酒,综合利用价值很大[5],但迄今为止,对爬山虎的研究甚少,对于爬山虎籽中总黄酮的研究尚未见报道。本文对爬山虎籽中黄酮的定性、提取工艺及含量测定进行了研究,以期为爬山虎籽的开发利用提供依据,为爬山虎的综合开发奠定基础。

    1 仪器与试药

    1.1 仪器

    60-CRT双光束紫外可见分光光度计(上海精密科学仪器有限公司);800型离心沉淀机(上海手术器械厂);HH-S型温水浴锅(江苏金坛市医疗仪器厂);pHS-3C精密pH计(上海雷磁仪器厂)。
