移植干细胞治中风:前景看好

     路透社健康消息 在最近召开的美国中风协会第26届国际中风会议上,一份在大鼠身上做的研究结果指出,把干细胞直接移植到脑部有可能修复被中风破坏的脑组织。

    干细胞属原始细胞,能在人体内分化成各种不同类型的细胞。医学界认为,干细胞在许多细胞被破坏的疾病的治疗上有着美好的前景,其中包括帕金森氏病和糖尿病。

    纽约艾伯特·爱因斯坦医学院的研究员报告说,他们用诱导的方法使成年老鼠的脑组织发生中风样改变,然后分别将干细胞移植到被诱导的老鼠和正常老鼠的脑部,进行观察对比。这些干细胞是从脑组织的皮层收集而来的,并经过荧光染色处理,在荧光显微镜下能发光,因此研究员可以追踪移植后干细胞的情况。

    研究发现,干细胞的独特之处在于它们在初期彼此十分相似,但当被移植到不同的部份后,它们就会分化成高度特异性的细胞,有着不同的功能。当被移植的干细胞分化成具有正常外形的脑细胞时,就叫做神经元。而且在一周内这些神经元会分化成与其所在区域功能相匹配的高度特异的细胞,充份成熟需要21至45天。

    研究员相信,终有一天这种细胞移植的方法可以用于替换被中风破坏的细胞,从而治疗中风病人。但在受损或有瘢痕的脑组织中生长的细胞并不象在正常脑组织中生长的细胞一样健康,这种生长中的改变可能是由瘢痕组织或其他因素造成的。因此在继续实验之前,有必要对影响移植细胞的因素有一个更好的了解。

    Cell Transplants May Heal Stroke-Damaged Brains

    FT. LAUDERDALE, Fla. (Reuters Health) - It may be possible to repair stroke-damaged brains by transplanting stem cells directly into the brain, according to results of a study in rats reported here at the American Stroke Association‘s 26th International Stroke Conference.

    Stem cells are primitive cells that have the ability to mature in many different types of cells in the body. They are believed to hold great promise in treating a number of ailments in which cells are damaged or destroyed, including Parkinson‘s disease (news - web sites) and diabetes.

    Researchers from the Albert Einstein College of Medicine in New York report that they transplanted stem cells into the brains of adult rats in which they had induced stroke-like damage. Drs. Gaurav Gupta and Daniel M. Rosenbaum also transplanted cells into normal rat brains. In both cases the cells flourished.

    The researchers ``harvested‘‘ stem cells from the outer layer of the brain. The cells were then treated with a fluorescent dye, which glows when viewed with a fluorescent microscope. This procedure permits the investigators to track the progress of the transplanted cells, Rosenbaum explained.

    Stem cells, Gupta noted, are unique in that they start out very similar to each other but then transform into highly specialized cells with very different functions depending upon where they are transplanted.

    Rosenbaum said that the transplanted cells developed into normal-appearing brain cells called neurons. Moreover, he said that in a week the cells were taking on very the highly specialized functions of the brain areas where they were transplanted. By 21 to 45 days, the cells were fully mature.

    Rosenbaum said it might some day be possible to use such cell transplants to treat stroke patients by replacing the cells damaged by stroke.

    But Gupta, who won an American Stroke Association award for his research, said that the cells do not grow as well in damaged or scarred areas of the brain as they do in healthy areas. He said this difference in growth might be due to the scar tissue itself or some other factor.
