Abstract
AIM: To investigate the distribution and effect of recipient-derived bone marrow mesenchymal stem cells (BM-MSCs) after orthotopic liver transplantation.
METHODS: The model of orthotopic liver transplantation was established using two-cuff method. Wistar rats were used as donors while Sprague Dawley rats were selected as recipients. The BM-MSCs were isolated by the combination of gradient density centrifugation and adhesion separation. The collected BM-MSCs were labeled with 5, 6-carboxyfluorescein diacetate succinimidyl ester (CFSE) and then infused into livers via portal vein after the operation. The distributions of BM-MSCs in liver, spleen, lung, and kidney were detected by fluorescence microscope 1, 2 weeks and 1 month after the operation. Hepatic functions and immunological rejection in liver tissues were also detected.
RESULTS: BM-MSCs collected by gradient density centrifugation grew in an adhesion mode on the wall and had a ball-like appearance ......
