重组BCG-Sj26GST疫苗诱导小鼠sIL-2R和IFN-γ的变化

    免疫学杂志010103 摘 要:目的 研究日本血吸虫重组BCG-Sj26GST疫苗对小鼠脾细胞sIL-2R和IFN-γ的影响。方法 第1次实验采用1×106和1×108CFU疫苗皮下免疫BALB/C鼠,免疫后8周用日本血吸虫尾蚴进行攻击感染,感染后6周剖杀小鼠,同时设有PBS对照组。第2次实验用1×106CFU疫苗皮下和静脉注射分别免疫小鼠,于免疫后0、4、8、10、14和16周各剖杀4只,分离脾脏,用Sj26或ConA刺激脾细胞,用ELISA法检测脾细胞上清液中sIL2-R和IFN-γ含量。结果 疫苗免疫,尾蚴攻击后,sIL-2R和IFN-γ水平显著升高;动态观察发现sIL-2R和IFN-γ分别于免疫后8~10周和4~8周达最高水平。结论 日本血吸虫重组BCG-Sj26GST疫苗可加强宿主Th1反应,促进Th1细胞分泌IL-2和IFN-γ,它们与免疫细胞相互作用,提高宿主抗血吸虫感染的保护力。azo, http://www.100md.com

    Abstract:Objective To investigate the effects of recombinant BCG-Sj26GST vaccine of schistosoma japonicum on sIL-2R and IFN-γ produced by spleen cells of mice. Methods In the first experiment, BALB/C mice were immunized subcutaneously by 1×106 and 1×108 CFU vaccine respectively, challenged with Sj cercariae 8 weeks after immunization and killed 6 weeks after infection to separate spleens, PBS served as control; in the second experiment, after immunized subcutaneously and intravenously by 1×106 CFU vaccine respectively,4 mice were killed to separate spleens on 0、4、8、10、14 and 16 weeks after immunization, spleen cells were stimulated by Sj26 or ConA. Levels of sIL-2R and IFN-γ in supernatant of spleen cells were detected by ELISA.Results Levels of sIL-2R and IFN-γ increased obviously after immunization with the vaccine against challenge with Sj cercariae; dynamic observation showed sIL-2R and IFN-γ reached the highest level on 8-10 weeks and 4-8 weeks after immunization respectively. Conclusion Recombinant BCG-Sj26GST vaccine of schistosoma japonicum may strengthen host’s Th1 response, promote Th1 cells to secrete IL-2 and IFN-γ. These cytokines and immune cells may influence each other to increase host’s protection against schistosomiasis.
