【摘要】 目的 探讨流式细胞术检测外周血淋巴细胞亚型的条件，确定流式细胞术检测淋巴细胞亚型样品的有效保存时间。方法 采集健康人外周血，4℃保存，在不同时间标记荧光抗体并对淋巴细胞亚型进行定量分析;将标记荧光抗体的样品用1%多聚甲醛固定并保存于4℃冰箱，于不同时间进行检测。结果 外周血样品保存至第10天，CD3 + 、CD4 + 、CD8 + 细胞数下降有显著意义(P<0.05，第18天P<0.01)，而CD4 + 、CD8 + 细胞比值变化无显著意义(P>0.05);固定样品在1个月内的各项指标变化无显著意义(P>0.05);裂解的最佳时间为25～30min。结论 保存于4℃的外周血样品在8天内检测结果差异无显著性，而固定样品在1个月内检测结果差异无显著性，这对于临床诊断和基础研究具有很大的实际意义。
