缺氧诱导促红细胞生成素在卵巢癌细胞系中的表达

     【摘要】 目的 探讨促红细胞生成素(Erythropoie,Epo)在卵巢癌细胞系中的表达及缺氧对其表达的影响,为进一步研究Epo在卵巢癌的发生发展中的作用打下基础。 方法 应用免疫细胞化学S-ABC法检测Epo蛋白、原位杂交法检测Epo mRNA在卵巢癌细胞系3AO中的表达及不同时间的缺氧处理后表达强度的改变。 结果 Epo及其mRNA在体外培养的卵巢癌细胞系中存在弱阳性表达,缺氧处理后表达明显增强,二者差异有非常显著性(P<0.001),Epo的表达主要定位于细胞浆、核膜。 结论 Epo在卵巢癌细胞系中存在基础表达且受缺氧影响,Epo可能在卵巢癌的恶性生长及抗凋亡等环节发挥作用。

    The expression of hypoxic-induced erythropoietin in ovarian carcinoma cells line

    【Abstract】 Objective To investigate the expression of erythropoietin(Epo)in normal and hypoxia in ovarian carcinoma cell line and to study the role of Epo in the tumorigenesis and development of ovarian carcinoma.Methods Immunocytochemical assay(S-ABC)was used to detect the expression of Epo in ovarian carcinoma cell line and to find the change of the intensity of the expressions by dealing with different hypoxic degree.Results The expression of Epo mainly locate in cytoplasm and karyotheca of ovarian carcinoma cells.The positive expression of Epo in normal was significantly lower than that in hypoxia(P<0.001).Conclusion The expression of Epo in ovarian carcinoma can be affected by hypoxia and Epo may play important roles in the anti-apoptosis and malignant development of o-varian carcinoma. ......
