EGb761对β-淀粉样蛋白损伤的保护作用

     【摘要】 目的 观察银杏叶提取物EGb761体外抗β-淀粉样蛋白(Aβ)损伤的作用。 方法 用MTT法检测细胞的增殖活性;释放到细胞外的LDH活性检测细胞膜的损伤;Caspase-3活性检测细胞凋亡。 结果 EGb761呈剂量依赖性提高Aβ损伤引起的SY5Y细胞的存活率降低,降低Aβ损伤引起的LDH释放及Caspase-3活性增加。 结论 EGb761对Aβ 25-35 损伤有保护作用。

    Effects of EGb761on SH-SY5Y cells against damage induced by Aβ

    【Abstract】 Objective To investigate the protective effects of EGb761on SH-SY5Y cells against damage induced by amyloidβ-peptide(Aβ).Methods The viability of SH-SY5Y cells was measured byMTT assay.The loss of membrane integrity and cell lysis was measured by assay the release of lactic dehydrogenase(LDH).The activ-ity of caspase-3in cells was measured by means of the cleavage of fluorogenic substrate Ac-DEVD-MCA.Results EGb761could respectively improve the survival of SH-SY5Y cells.The results also showed that EGb761signifi-cantly decreased Aβ-induced LDH release and reduced the Aβ 25-35 -induced increase of caspase-3activity.Con-clusion EGb761protected SH-SY5Y cells against toxicity induced by Aβ. ......
