HGPRT基因缺陷型骨肉瘤细胞系LM9的建立及其生物学特性测定

     【摘要】 目的 建立次黄嘌呤鸟嘌呤磷酸核糖基转移酶(HGPRT)基因缺陷型骨肉瘤细胞系LM9,进行细胞形态、生长特性及致瘤性等生物学特性检测。 方法 应用N-甲基-N′-硝基-N-亚硝基胍(MNNG)诱导骨肉瘤细胞系LM8突变,用8-单杂鸟嘌呤(8-AG)筛选HGPRT基因缺陷型骨肉瘤细胞系LM9;观察细胞形态、细胞核型分析、细胞生长周期等生物特性,接种C3h鼠观察致瘤性。 结果 突变后细胞仍具有典型的成骨肉瘤细胞特征,细胞异型性明显,染色体众数为64-66,接种鼠具有成瘤性,成瘤率为100%。 结论 本实验建立了HGPRT基因缺陷型骨肉瘤细胞系LM9,具有典型的鼠源性骨肉瘤细胞特征,可用于杂交瘤、肿瘤疫苗等多方面的研究,是一个较好的实验工具。

    【Abstract】 Objective To establish the HGPRT-defect osteosarcoma cell lineage,and determine the ability of sarcomagenesis and fusion.Methods N-methyl-N′-nitro-N-nitrosoguanidine(MNNG)was used to induce the mutation of an osteosarcoma cell lineage(LM8),then culturing mixed with8-azaguanine killed those cells of HG-PRT positive,We got the HGPRT-defect osteosarcoma cell lineage LM9.The LM9cells were observed on cells biolo-gy characteristic such as morphous and cell cycles biology inoculated into mice and their growths were surveyed in the mice.Results The LM9cell lineage has typical characteristics of osteosarcoma cells and it is sarcomagenic to C3h mice.Conclusion The LM9cells lineage was obtained by method of mutation ......
