【摘要】 目的 研究和比较跨膜型肿瘤坏死因子-α(transmembrance TNF-α，mTNF)和分泌型TNF-α(secreted TNF-α，sTNF)对细胞毒T淋巴细胞(cytotoxic T lymphocyte，CTL)功能的影响。 方法 采用单向混合淋巴细胞培养的方法，获取针对同种异体MHC分子的CTL。 结果 相同的刺激细胞经ConA刺激后作为靶细胞，成功建立了CTL实验模型;混合培养白细胞(mixed leukocyte culture，MLC)，与靶细胞作用后，两型TNF-α均有表达:mTNF-α表达增加约2倍(P<0.05)，sTNF-α增加3倍(P<0.05);MLC与抗TNF-α抗体预先作用，再加入靶细胞，该抗体不能有效阻断其杀伤作用;相同剂量两型外源性TNF-α与MLC作用24h后，对MLC的杀伤作用不明显。 结论 两型外源性TNF-α对MLC作用差异无显著性。

    Effect of secreted and transmembrane tumor necrosis factor-αon the functions of human MLC

    【Abstract】 Objective To study and compare the effect of mTNF-αand sTNF-αon the cytotoxic function of CTL.Methods CTL specific for allogenic MHC was taken by one-way mixed lymphocytes culture.Results Ex-perimental model of CTL used as target cells was succeeded stimulated by ConA.The two forms of TNF-αwere ex-pressed after the MLC reacted with their target cells.The mTNF-αelevated by2folds(P<0.05)vs sTNF-αele-vated by3folds(P<0.05).After primed with the antibody of TNF-αand reacted with their target cells the anti-body could not block the cytotoxicity of MLC.MLC were challenged with the two forms of exogenous TNF-αin samedose.Conclusion The cytotoxic effect of the two forms of TNF-αon the MLC is inapparent. ......
