提 要 目的:了解重组RA538、反义c-myc腺病毒载体对正常细胞的影响。方法:采用形态学观察、MTT、RT-PCR等方法,对腺病毒介导RA538及反义c-myc基因对正常人胚肺成纤维细胞(2BS)的影响进行研究。结果:以Ad-LacZ进行重组腺病毒转导效率检测发现腺病毒2BS细胞具有很高的转导效率。Ad-RA538、Ad-ASc-myc对正常人胚肺2BS细胞无生长抑制及凋亡诱导作用,同时Ad-RS538对2BS内源性c-myc、bcl-2基因的表达也无影响。结论:腺病毒载体具有很高的转导效率,能实现目的基因在转导细胞中的高水平表达。Ad-RA538、Ad-ASc-myc对2BS细胞无明显的生长抑制及凋亡诱导作用,对2BS细胞内源性bcl-2、c-myc基因的表达无调节作用。

Effects of recombinant RA538 antisense c-myc adenovirus on human normal cells

Abstract Objective: To study theeffects of recombinant RA538 antisense c-myc adenovirus (Ad-ASc-myc) on 2BS human normalcell line. Methods: 2BS human normal cells were treated with Ad-RA538, Ad-ASc-myc and Ad-LacZ and morphological changes and others were observed and MTT nad RT-PCR wereemployed. Results: Ad-RA538 and Ad-ASc-myc could not inhibit the growth of 2BS cells orinduce their apoptosis. Ad-RA538 could not affect the expression of endogenous genes of c-myc and bcl-2. Conclusion: Ad-RA538 and Ad-ASc-myc can not inhibit the growth or induceapoptosis of 2BS cells and can not down regulate the expression of bcl-2 and c-myc geneson 2BS cells.
