荷瘤小鼠口服醋酸铜对器官组织铜与锌动力学的影响

摘要 目的:观察荷瘤小鼠体内各器官组织铜锌的变化及口服醋酸铜(150 mg/kg)后铜、锌在体内的动态分布过程。方法:用Balb/C小鼠造成实体瘤模型,用原子吸收方法测定组织中的铜锌含量。结果:荷瘤小鼠肝脏及胃肠组织的铜水平明显低于正常对照组(均P<0.01),血清及脾脏组织高于对照组(均P<0.01)。与此同时心脏组织及血清的锌含量较正常对照组出现有意义的下降(均P<0.05),而肝脏、脾脏则出现有意义的升高(P<0.05,P<0.01)。荷瘤小鼠口服铜盐后,所观察的组织中先后出现不同程度的升高。铜高值出现在0.5 h的是血清;到2 h的有心脏、脾脏、肺脏、肾脏、胃肠,而5 h时血细胞、肿瘤组织出现最高值;肝脏组织的铜在12 h时仍在上升;锌随着铜的变化而变化,锌高值出现的时间一般与铜出现高值的时间同步或后移(除肝脏)。结论:荷瘤使机体内铜锌分布发生变化,血清铜升高、肝脏及胃肠等重要组织中的铜降低。给铜后,荷瘤小鼠体内铜、锌的动态变化因组织不同而异,肝脏是铜聚集的主要器官。

KINETIC CHANGES OF COPPER AND ZINC

    IN TUMOR BEARING MICE DURING COPPER LOAD

Abstract Objective:To observe the changes of copper and zinc in tumor bearing mice during tumor development and copper loaded.Methods:Atom absorptive spectrometry was used to analyse the kinetic changes of copper and zinc in different tissues of tumor bearing mice.Results:The copper level in tissues of liver and stomach-intestine of the bearing tumor group(BTG)was greatly lower than that in the normal control group(CG) respectively(P<0.01),whereas in serum and spleen,the copper level was higher in BTG than that in CG(P<0.01).In lung,heart,kidney and blood cell,there was no significant difference between both the groups.The zinc level in heart and serum of BTG were significantly decreased than that in CG(P<0.05),but in liver and spleen zinc was increased than that in CG(P<0.05,P<0.01 respectively).After oral administration of copper acetate,the highest level of zinc appeared at the same time or some time later with the copper changes in all other tissues than liver tissue.In tumor tissue,the basic value of copper and zinc were markedly lower than that in other tissues than serum and blood cell.Conclusion:Serum copper was increased while the copper in liver and stomach-intestine was markedly decreased.After copper loaded,distribution of copper and zinc is changed in different tissues.Among them,liver is the major tissue accumulated more copper than other tissues and appeared strong toleration to copper.
