α-干扰素对膀胱癌细胞多药耐药性逆转的研究

摘要 目的 研究α -干扰素(α -IFN)对膀胱癌多药耐药性的逆转作用。 方法 应用 MTT法检测阿霉素(ADM)的细胞杀伤作用。荧光法检测细胞内阿霉素浓度和斑点杂交法检测mdr1 mRNA相对含量。 结果 500 IU/ml α IFN能明显增强阿霉素对耐药细胞BIU-87/ADM的杀伤作用,但不能增强阿霉素对敏感细胞BIU-87的杀伤作用;α -IFN能增加阿霉素在BIU-87/ADM细胞内的积聚,但不改变mdr1 mRNA在细胞内表达量。 结论 α -IFN对膀胱癌细胞多药耐药性具有逆转作用,其机制与α-IFN增加细胞内抗癌药物浓度有关,与增加mdr1 mRNA表达量无关。

Abstract Objective To study the reversal of multidrug resistance by α -IFN in bladder cancer cell line. Methods The effect of recombinant human α -interferon (α IFN) on the cytotoxicity of adriamycin(ADM) in human bladder cancer cell line BIU 87 and its multidrug resistant counterpart BIU-87/ADM were studied by MTT assay. ADM accumulation and level of mdr1 mRNA in above cell lines were also detected by spectrofluometric and dot blot analysis respectively. Results α -IFN (500IU/ml)increases the cytotoxicity effect and the accumulation of ADM in BIU-87/ADM cell line,but had no effect on the cytotoxicity of ADM to BIU-87 cell line,the level of mdr1 mRNA being unchanged. Conclusions α -IFN has significant effects on the reversal of multidrug resistance of human bladder cancer cell line.
