摘要 为探讨多肿瘤抑制基因1(MTS1)在原发性膀胱癌发生发展中的作用,采用PCR-SSCP和DNA直接测序方法,对18例膀胱移行细胞癌进行MTS1分析。结果18例原发性膀胱癌中没有发现MTS1纯合性缺失,但发现2例点突变。表明原发性膀胱癌的发生与MTS1的失活相关。

Abstract Abstract Multiple tumor suppressor 1(MTS1), encoding a Mr16 000 protein(p16) ,maps to chromosomal 9p21. The p16 protein inhibit the kinase activity of cyclinD/CDK4 complex and blocks its pregression to S phage, thus inhibiting cell growth anddivision and blocks the malignant transformation of cell. When p16 gene was inactivatedresulting from homozygous deletion or point mutation, its function as a tumor inhibitorwill be lost,resuling in tumor development. To elucidate the role of MTS1 gene intumorigenesis of bladder cancer,18 cases of bladder cancer were analyzed by polymerasechain reaction(PCR),PCR-single strand conformational polymorphism (SSCP) and DNAsequencing. No homozygous deletions were found in the MTS1 gene,while point mutations werefound in 2 of 18 bladder cancers. It was concluded that tumorigenesis of primary bladdercancer is correlated with inactivation of MTS1 gene.
