【摘要】 目的 建立多药耐药细胞系。 方法 将带有mdr1 cDNA全序列的逆转录病毒载体pHaMDR通过脂质体直接导入到敏感的人膀胱癌细胞系BIU-87细胞中。 结果 经60~100ng/ml秋水仙素筛选21天后,克隆出高强度MDR表型的耐药细胞系,基因组DNA PCR扩增表明mdr1 cDNA已掺入到BIU-87细胞基因组中。 结论 应用转基因方法可建立多药耐药细胞系,该细胞系具有耐药强度高,耐药性稳定等特点。

Construction of a multidrug resistant cell line with gene transfer

【Abstruct】 Objective To construct a multidrug resistant cell line. Methods The retroviral plasmid pHaMDR that contains the full sequences of mdr1 cDNA has been transfected into BIU-87 cell line, a sensitive bladder cancer cell line, by liposome. Results A highly resistant cell line was astablished after cultured with selective medium containing 60~100ng/ml colchicine for 21 days. The PCR results confirmed that mdr1 cDNA had been transduced into the genome of the sensitive bladder cell line. Conclusions Gene transfer is a sound method to construct multidrug resistant cell line. The transfected cell line showed high degree and stability of multidrug resistance.
