【摘要】 目的 研究氟化钠、甲基汞对骨组织的作用,以探讨其致畸性。方法 将氟化钠或甲基汞直接加入培养基中,剂量分别为:0、10、20、30或0、0.10、0.25、0.50 mg/L,观察培养后肢体的发育和分化情况,并用图像分析仪测量肢体中各软骨原基的面积和形状。结果 (1)随着染氟量的增加,肢体中多数软骨原基的发育和分化越差,面积越小,并对软骨原基的外形发育产生抑制。(2)随着染甲基汞量的增加,肢体中各软骨原基的发育和分化越差,面积越小,并对软骨原基的外形发育产生抑制,爪骨受到的影响比长骨严重,对骨组织的作用比对软组织大。结论 过量的氟化钠或甲基汞具有潜在的致畸性。

Application of the mouse limb bud culture to study the teratogenicity of fluoride or methylmercury

Abstract Objective In this paper the teratogenic potential of fluoride and methylmercury were studied. Methods An in vitro method of mouse limb bud culture in self-made rotator with continuous supplem entation of gas mixture was used.Image analysis of the area and the form of the bone anlagen of the cultured limb was used to evaluate quantitatively their teratogenic potentials.Fluoride(0,10,20,30 mg/L medium) or methylmercury(0,0.10,0.25,0.50 mg/L medium) was added to limb bud cultrues of 12 day-old mouse embryos (Kunming Strain). Results After 3 day incubation,as the dose increased,the area of the bone anlagen oflimbs cultures decreased significantly and the form factors increased.The effects were more severe in the caseof methylmercury than fluoride.Methylmercury had a greater effect on chondrogenic tissue than on softtissue,and on the paw bone than on long bone.
