摘要 目的 研究大鼠脑缺血-再灌注(CIRI)时热休克蛋白(HSP)70表达及川芎嗪对HSP70表达的影响。方法 采用大鼠左侧颈总动脉结扎并滴注生理盐水脑CIRI模型,以免疫细胞化学法检测24只鼠脑CIRI及川芎嗪干预CIRI脑组织HSP70表达,并与病理组织学改变进行对照研究。结果 (1)非缺血侧脑组织无HSP70表达,缺血30分钟缺血侧大脑皮层可见HSP70表达;再灌注30分钟组仅1只大鼠(1/6)缺血侧大脑皮层有HSP70表达。(2)与单纯缺血大鼠相比,川芎嗪干预组缺血侧大脑皮层HSP70表达明显增强,计算机辅助图像半定量分析,HSP70免疫阳性细胞数,分别为平均43.6±12.5个/片和85.0±16.7个/片,两组间差异非常显著(P<0.001)。(3)缺血侧皮层神经细胞呈现轻度缺血改变,部分细胞核固缩深染,细胞缩小,细胞周围间隙增大;川芎嗪干预大鼠缺血损伤程度有减轻,细胞核固缩减轻,细胞无缩小。结论 脑缺血时可诱导缺血脑细胞部分HSP70表达,川芎嗪干预可使缺血大脑皮层HSP70表达明显增强,皮层神经细胞的缺血损伤减轻。

Abstract Objective To study the expression of HSP70 and the effects of tetramethylpyrazine (TMP) on cerebral ischemic reperfusion injury (CIRI) in rats. Methods By using an experimental cerebral ischemia model in 24 Sprague-Dawley (SD) rats, the cerebral cellular expression of HSP70 and the effects of TMP, a blood-activating and stasis-eliminating herb, on CIRI were studied by immunohistochemical staining in conjunction with histopathological analysis of rat brain. Results (1) No HSP70 was found in non-ischemic brain tissue but HSP70 was observed in the parietal cortices 30 minutes after brain ischemia. But, 30 minutes after reperfusion, no HSP70 was observed except in one of the six rats with brain reperfused. (2) As compared with that of cerebral ischemia rats, HSP70 expression increased significantly and the number of HSP70 immunoreaction positive cells in cerebral ischemia rats treated with TMP increased markedly as shown by computer image analysis (from 43.55±12.51/section to 84.95±16.70/section, P<0.001). (3) After cerebral ischemia, most neurons were found morphologically intact except some mild cerebral ischemic changes in the parietal cortex, such as shrinkage of neuron and nucleolus and smearing of nucleolar-cytoplasmic border. In the TMP group, though some mild ischemic changes in the parietal cortex still existed, they were alleviated with no neuron and only slight nucleolus shrinkage. Conclusion Cerebral ischemia induces HSP70 protein in the injured cells and the administration of TMP increases HSP70 expression and alleviates cerebral ischemic changes histopathologically.
