【摘要】 目的 从细胞和基因水平阐明大蒜油对人胃癌BGC-823细胞的作用机理。方法 用激光扫描共聚焦显微镜及Northern杂交等技术进行分析。结果 大蒜油对癌细胞形态、增殖和致瘤性有明显影响。结论 大蒜油通过促进p53、p21等的表达,诱导肿瘤细胞分化,同时诱导肿瘤细胞凋亡。

    Garlic oil induces differentiation and apoptosis of human gastric cancer cell line Li Xiaoguang, Xie Jinyu, Li Wenmei, et al. China Rehabilitation Research Center, Beijing100077

    【Abstract】 Objectives To study the mechanism of garlic oil-induceddifferentiation of human gastric cancer cell line BGC-823. Methods Confocal microscopicassay and Northern blot hybridization were used to detect morphologioal change and geneexpression in garlic oil treated human gastric cancer cells (BGC823). ResultsCell growthin vitro and tumorigenicity in nude mice were inhibited dramatically by garlic oil. Garlicoil treatment resulted in cell apoptosis as shown by DNA fragmentation. High expressionlevel of WAF1/p21 and wild type p53 gene was detected in garlic oil-treated cells.Conclusion These data indicate that differentiation and apoptosis of BGC-823 cell can beinduced with garlic oil, and p21 and p53 genes possibly play important role in theprocess.
