【摘要】 目的 探讨人工合成杀菌肽(Shiva-1)对肿瘤细胞的杀伤及选择性杀伤作用机制。方法 采用3H-TdR掺入法,细胞内游离钙含量及动态变化测定。扫描、透射电子显微镜法,观察人工合成杀菌肽Shiva-1对人肝癌细胞的生长抑制、细胞DNA合成、细胞内游离钙离子含量的动态变化及对超微结构的影响。结果 人工合成杀菌肽Shiva-1可抑制癌细胞生长及3H-TdR掺入细胞DNA,使细胞内游离钙含量迅速降低,细胞超微结构改变;正常猴肾细胞CV1未见损伤。结论 人工合成杀菌肽Shiva-1对肿瘤细胞具有杀伤及选择性杀伤作用;作用起始部位在细胞的膜性结构;作用机制不同于化学抗癌药物。细胞内游离钙含量降低可能与其选择性杀伤作用有关。

    

CYTOCIDAL EFFECT OF ASYNTHETIC CECROPIN SHIVA-1 ON LIVER CANCER CELL

【Abstract】 Objective To elucidate the cytocidal effect andselective cytocidal effect mechanism of synthetic cecropin Shiva-1 on the carcinoma cells.Methods 3 H-TdR incorporation, measurements of intracellular Ca2+with the ACAS 570 (Adherent Cell Analysis and Sorting), and electron and light microscopywere used to observe the effects of synthetic cecropin Shiva-1 on the liver cancer cellsBEL-7402. Results The synthetic cecropin Shiva-1 inhibits the proliferation andincorporation of 3 H-TdR into BEL-7402 cells at a lower concentration (5μ g/ml). The action is time-andconcentration-dependent. The supermicro-structures of BEL-7402 cells changed after acting30 minutes with synthetic cecropin Shiva-1. By 24~ 48 hours, the membranes of the carcinoma cells had beendamaged and the cells had been lysed. The levels of intracellular Ca 2+ decreasedrapidly within 50 seconds after adding the cecropin Shiva-1. The quantity of intracellularCa2+ of BEL-7402 cells decreased significantly than normal mammal cells (CV1) andcarcinoma cells without cecropin Shiva-1 action. This damage could not be observed innormal mammal cells (CV1). Conclusion The synthetic showed selective cytocidal effect onthe carcinoma cells but no damage to normal mammal cells. The initial acting area waslocated on the membranous structure. Thus the permeability of cellular membrane wasincreased, potential difference and osmotic integrity was lost. This mechanism differedfrom chemical anticarcinogen. These results suggested that synthetic cecropin Shiva-1might be an effective therapeutic agent for human carcinoma.
