摘要 目的 了解16型人乳头瘤病毒(HPV16)E6/E7、HSP70与角质形成细胞永生化及转化之间的关系。方法 Lipofectamine介导法进行转染,以G418筛选阳性克隆,免疫印迹分析及免疫荧光双标记染色结合激光扫描共聚焦显微镜对HSP70和HPV16E6/E7在转染角质形成细胞中的表达进行了观察。结果 免疫印迹分析显示正常角质形成细胞和转染角质形成细胞均在相对分子质量70000处出现一条染色带,但转染细胞的着色深度明显强于正常细胞;激光扫描共聚焦显微镜观察也发现转染细胞中HSP70的表达明显增高,HSP70与HPV16E6/E7有共定位。结论 HPV16E6/E7可以诱导角质形成细胞中HSP70的高表达,这种高表达可能在角质形成细胞永生化甚至转化过程中具有重要作用。

Expression of 70-kD Heat ShockProtein in Keratinocytes Induced by Human Papillomavirus 16 E6/E7

Abstract Objective To investigate the relation between human papillomavirus16 E6/E7 (HPV16 E6/E7),70000 heat shock protein (HSP70) and immortalization andtransformation of keratinocytes. Methods Keratinocytes were transfected with lipofectamineand positive clones were selected by G418. The expressions of HSP70 and HPV16 E6/E7 bytransfected keratinocytes were analysed by means of Western blot,double labellingimmunofluorescent staining and laser -scanning confocalmicroscopy techniques. Results Results of Western blot analysis showed a specific bandexhibiting a molecular weight of 70 000 in both normal keratinocytes and HPV16 E6/E7transfected keratinocytes, but the staining was more intensive in transfectedkeratinocytes than in normal ones. The co -location ofHPV16 E6/E7 and HSP70 in transfected keratinocytes were observed by laser -scanning confocal microscopy. Conclusion The high expression ofHSP70 in transfected keratinocytes may be induced by HPV16 E6/E7 which suggests that HSP70might play an important role in immortalization and even transformation of keratinocytes.
