摘要 目的 探讨外周T淋巴细胞功能活性与Alzheimer病(AD)病情的关系。方法 流式细胞仪测定T细胞亚群;MTT法检测ConA刺激AD外周淋巴细胞的增殖功能;ELISA法检测细胞因子及其受体表达。结果 (1)CD4+、CD8+T细胞比例和CD4+ /CD8+ 比值各组间无统计学差异。(2)淋巴细胞增值在轻、重AD组略下降,但刺激指数(SI)都大于2,与对照无明显差异。(3)IL-2的分泌在重度AD组和sIL-2R表达在轻、重度AD组明显升高(P<0.05),IL-2R的表达各组间差异无显著性。结论 T淋巴细胞亚群变化与AD病情的联系不明显,细胞因子分泌功能与AD进展有关。

Immune-associated Function and Activity of T Lymphocyte in Alzheimer Disease

Objective To investigate the relationship between the immune-associated function and activity of Tlymphocyte and severity of Alzheimer s disease stage. Methods The patientswere divided into two groups-mild and moderately severe-according to severity of diseasestage, and their lymphocytes were compated to those of elderly controls. Monoclonalantibodies labeled by FITC were used to lymphocyte subset analysis. Lymphocytes werestimulation with ConA, we observed the proliferative ability, IL-2 secretion, cellmembranal IL-2R expression and sIL-2R expression. Results 1. Nosignificant differences were found in T lymphocyte subpopulations and the ratios ofCD4/CD8 between groups (P > 0.05). 2. All groups revealed strong response to ConA, there were no significantdifferences between AD and controls (P > 0.05). 3. The IL-2 levels showed increase in AD groups andthere was significant differences between moderately severe AD and controls (P < 0.05).The amount of sIL-2Rrevealed an increase in AD groups, there were significant differences between AD andcontrols (P < 0.05).The expression of IL-2R revealed no significant differences between AD and controls(P > 0.05). Conclusions Our results indicated that immunologic parameters may be useful in monitoring progressionof AD.
