脱蛋白骨、骨形态发生蛋白和三种生长因子重组合人工骨成骨能力的比较研究

【摘 要】 目的 比较三种重组合人工骨的成骨能力。方法 在50只兔的100个颅骨缺损动物模型中分别植入:①牛脱蛋白骨(bDPB)/牛骨形态发生蛋白(bBMP)/肿瘤坏死因子α(TNFα);②bDPB/bBMP/碱性成纤维细胞生长因子(bFGF);③bDPB/bBMP/表皮细胞生长因子(EGF);④bDPB/bBMP。植入后1、2、4、6及8周行组织学检查及X线摄片;植入后10及42天行 35 S和45 Ca液闪计数及灰重测定。结果 成骨能力:bDPB/bBMP/TNFα>bDPB/bBMP/bFGF(P<0.01)>bDPB/bBMP/EGF(P<0.01);bDPB/bBMP/EGF与bDPB/bBMP无显著性差异(P>0.05)。结论 TNFα与BMP及载体复合后,在体内对骨修复产生明显的促进作用,在载体/骨诱导因子/生长因子的重组合人工骨模式中,DPB/bBMP/TNFα是一种有价值的骨移植材料。

COMPARATIVE STUDY ONOSTEOGENESIS OF THREE TYPES GROWTH FACTORS RECOMBINATION ARTIFICIAL BONES

Abstract Objective To compare the osteogenesis of recombination artificialbones, which are bovine deproteined bone (bDPB) and bovine bone morphogenetic protein (bBMP), combined with tumor necrosis factor α (TNFα ),basic fibroblast growth factor (bFGF), epidermal growth factor (EGF) respectively.Methods One hundred trephined skull bone defects in fifty rabbits weredivided into four groups, which implanted with bDPB/bBMP/TNFα ,bDPB/bBMP/bFGF, bDPB/bBMP/EGF, and bDPB/bBMP respectively.X-ray and histological changes were observed in the 1st, 2nd, 4th, 6th, 8thweeks after implantation.The content of 35 S and 45 Ca and ash weight were measured at 10 and 42 daysafter operation.Results The osteogenesis of bDPB/bBMP/TNFα group was stronger than that of bDPB/bBMP/bFGF group(P< 0.01), while bDPB/bBMP/bFGF groupwas stronger than that of bDPB/bBMP/EGF(P< 0.01).No significant statistical difference were found between bDPB/bBMP/EGF and bDPB/bBMP(P> 0.05).Conclusion TNFα combined with bBMP and carrier can stimulate bone formation andincrease the volume of new bone in vivo.It suggests that bDPB/bBMP/TNFα is a valuable biomaterial of bonegraft.
