摘 要 目的 为探索一种以体外培养方法收获弓形虫速殖子的较适条件。方法 以RH、SH1、PP3个分离株的弓形虫速殖子,按105 、104 、103 的接种量分别接种HeLa细胞,调定培养基的pH值分别为8.0、7.0和6.5进行培养。结果 发现接种后6d速殖子的累积收获量存在差异性,其中以SE1株收获量大,RH株次之,PP株最少;接种量为105 时,收获量大,104 组次之,103 最少。在培养基pH为6.5时的收获量大,pH7.0次之,pH8.0最少。结论 pH6.5的培养条件较适于试验的3株弓形虫速殖子体外增殖,同时,弓形虫的体外增殖也与接种量及不同弓形虫分离株有关。

A STUDY ON IN-VITROCULTIVATION OF 3 STRAINS OF

    TOXOPLASMA GONDII TACHYZOITES

ABSTRACT Aim To investigate therelatively optimal conditions for in-vitro cultivation of T.gondii for obtaining a betterharvest of tachyzoites.Method 3 isolates,viz.RHSH1PP,were cultivted in HeLa-cells with 3 different iniitalinoculates(105 104 103 )and adjusted the medium in 3 pHs(8.07.06.5).Results It was found that,harvestingat the 6th day after inoculation,there were distinct differences among the cumulativeharvest of tachyzoites.Harvest collection in SH1 was the highest versus PP the lowest.Whenthe pH 6.5 was applied,the harvest was higher than that of 7.0,and in 8.0 was thelowest.Similarly,the harvest decreased along with decreases of the initial inoculum,i.e.105 ,thehighest and 103 ,the lowest.Conclusion It may be concluded thatpH 6.5 is a suitable environment for multiplication of T.gondii tachyzoites in-vitro.Theamount of the initial inoculum and the strain of isolate difference may also haveinfluence on multiplication of the parasite in-vitro.
