【摘要】 目的 研究人垂体腺瘤中雌激素受体基因(estrogenreceptor gene, ER) mRNA与蛋白水平的表达及其与垂体腺瘤生物学特性的关系。方法 用免疫组化法和逆转录-聚合酶链反应(RT-PCR法),检测60例各种类型垂体腺瘤标本中ER蛋白的表达,比较不同类型垂体腺瘤ER蛋白表达的差异及ER蛋白表达与mRNA表达的差异。结果 泌乳素腺瘤、促性腺激素细胞瘤中均有ER蛋白表达,其表达与mRNA表达一致;无功能腺瘤中2例有ER蛋白表达,其蛋白表达与mRNA表达不一致;其它类型垂体腺瘤未发现ER基因表达。结论 ER基因表达的检测可有助于评价PRL腺瘤的增殖活性及筛选抗雌激素治疗对象。ER基因的表达异常可能与PRL腺瘤及促性腺激素细胞瘤的发生、发展有关。

Proteinand mRNA expression of estrogen receptor gene in pituitary adenomas.

【Abstract】ObjectiveThedifference among estrogen receptorl (ER) gene expression in various kinds of pituitaryadenomas and the correlation between ER protein and mRNA expression were explored. It mayprovide a clue to the etiology of pituitary adenomas and lays a fundation for furtherresearch about ER gene mutation and antiestrogen therapy in pituitary adenomas. Methods ERgene expression were evaluated by immunocytochemistry and RT-PCR in 60 human pituitaryadenomas, and the correlation between ER protein and mRNA expression was analyzed. Theresults were compared with ademomas hormone secretion and some other biological index.Results All PRL adenomas and gonadotroph adenomas were found to have ER protein and mRNAexpression. ER protein expression correlated with mRNA expression. Some of the nonfunctionadenomas had ER gene expression, but their mRNA expressions were not correlated with ERprotein expression. Other types of pituitary adenomas did not have ER gene expression.Conclusions Detection of ER gene expression can be regarded as an index of proliferationof PRL adenomas. Antiestrogen therapy may be an adjunct therapy for PRL adenomas. Theabnormal of ER gene expression may account for PRL and gonadotroph adnomas’occurranceand development.
