【摘要】 目的 研究胃癌中SOX4基因的碱基及其编码的氨基酸突变情况，探讨其在胃癌发生中的作用。方法 提取胃癌肿瘤及癌旁组织的DNA，PCR扩增胃癌SOX4基因的HMG-box框，并对其进行PCR-SSCP分析，对有差异的样本进行DNA测序。将测序结果用ClustalX软件与正常的HMG-box框序列比对，用DNAStar软件将HMG-box框碱基序列转化成氨基酸序列，再用ClustalX软件与正常的氨基酸序列比对。 结果 经序列测定发现在27例胃癌样本中有12例出现点突变，而在癌旁组织中没有发现突变，晚期胃癌的突变率明显高于早期胃癌。 结论 SOX4基因突变可能与胃癌的发展和转移有相关性。本研究为探索SOX4基因突变与人类肿瘤发生之间的关系提供了分子资料。
