Abstract
AIM: To investigate the antiviral activity on Hepatitis B Virus andthe gene responses ofinterferon alpha with different subtypes in vitro, and to assessthe feasibility of using the signal transduction molecules as a newstandard for evaluating the antiviral activities of IFN-asubtypes.
METHODS: After 2.2.15 cells were respectively treated with IFN-a2b, IFN-a2a, IFN-a1b of varied concentrations (0.5,1,2, 4, 8 MU/L), the contents of HBsAgand HBeAg in the supernatant were measured by Abbot kit and the inhibitoryrates on HBsAg and HBeAg were calculated. After HepG2 cells were treatedwith 1MU/LIFN-a2b, IFN-a2a, IFN-a1b, the levels of STAT1, IFNAR mRNA and protein of were detected by reverse transcription polymerase chain reaction (RT-PCR)and Western blotting ......
