【摘要】 目的 通过对MHCC97肝癌细胞株的FAK基因的表达进行干预，观察MHCC97肝癌细胞侵袭转移能力的变化。 方法 用Oligofec TAMINE介导的方法，将反义FAK ODN引入MHCC97肝癌细胞株，或用Genistein抑制MHCC97肝癌细胞FAK磷酸化方法，观察经过不同干预后的MHCC97肝癌细胞穿透Matrigel胶处理后的Transwell小室的活力变化，比较MHCC97肝癌细胞转染反义FAK ODN前后和用Genistein抑制磷酸化前后，细胞侵袭转移能力的变化。 结果 反义FAK ODN转染的肝癌细胞和用Genistein干预的肝癌细胞穿过Matrigel胶的能力减弱。 结论 抑制FAK的表达能影响肝癌细胞的侵袭转移能力。

    【Abstract】 Objective To study the inhibitory effects of antisense focal adhesion kinase(FAK)oligodeoxynucle-otides(ODN)and Genistein on the invasion of MHCC97hepatocellular carcinoma cells.Methods Oligofec TAMINE-mediated antisense FAK ODN was transfected and Genistein was added into MHCC97hepatocellular carcinoma cells.The invasive activity of tumor cells was assayed in a transwell cell culture chamber has been dealed withMatrigel first.Compare the changes between them.Results Antisense FAK ODN and Genistein can both decrease the invasion of MHCC97hepatocellular carcinoma cells.Conclusion Inhibition of FAK expression can inhibit the invasion ability of MHCC97hepatocellular carcinoma cells. ......
