槟榔碱诱导血管内皮细胞凋亡Caspase-3活性的研究

     [摘要] 目的:初步探讨凋亡“核心蛋白酶”——Caspase-3在槟榔碱(Arecoline)诱导血管内皮细胞(Endothelial Cells, EC)凋亡中的作用。方法:体外培养人脐静脉内皮细胞(Human Umbilical Vein Endothelial Cells,HUVECs),将EC与槟榔碱共同培养,应用荧光显微镜观察EC凋亡情况,计算凋亡百分率并用比色法检测Caspase-3的活性。结果:槟榔碱诱导血管内皮细胞组在荧光显微镜下可见到明显的细胞凋亡改变,Caspase-3活性较正常对照组明显增高(P<0.05),且在12 h时最高。结论:槟榔碱可诱导血管内皮细胞凋亡,Caspase-3可能参与了这一细胞凋亡过程的调控。

    Study on the Caspase-3 Activity in the Arecoline-induced Apoptosis of Endothelial Cells.

    [Abstract]Objective: The purpose of the study is to investigate the mechanism of the change of caspase-3 activity on the arecoline-induced apoptosis in endothelial cells(EC). Methods: HUVECs were cultured in virto. Apoptosis of HUVECs were induced by arecoline. The changes of apoptotic cells and caspase-3 activity in EC were evaluated with the fluorescecet microscopy and the colorimeteric assay respectively and were compared with normal control group. Results: Obvious cell apoptotic change could be observed with fluorecent microscopy after arecoline stimulation; The level of caspase-3 activity was significantly higher in the arecoline group than that of normal control group(P<0.01). Conclusion: Arecoline may induce the apoptosis of HUVECs and caspase-3 takes part in the control of apoptotic process. ......
