     [摘要] 目的:探讨人基因重组骨形成蛋白-2(rhBMP-2)对牙周韧带细胞群(PDLC)成纤维表型的影响。方法:PDLC在含不同浓度的rhBMP-2的培养液中培养,采用免疫细胞化学技术做表皮生长因子受体(EGFR)的检测,并用图像分析软件分析EGFR表达的强弱。结果:rhBMP-2作用下,PDLC的EGFR的表达有变化(P<0.05),25~100 μg/L组EGFR的表达减弱,100 μg/L组最弱,随rhBMP-2浓度上升,EGFR的表达又有增强的趋势。结论:rhBMP-2对PDLC的成纤维表型产生了影响,合理剂量的rhBMP-2应用可以保证PDLC成纤维表型的较高表达,使种植体周牙周韧带的构建成为可能。

    Effects of Recombinant Bone Morphogenetic Protein-2 on Fibroblastic Phenotype of Periodontal Ligament Cells Population.

    [Abstract]Objective: To explore the effects of rhBMP-2 on the fibroblastic phenotype of PDLC. Methods: PDLC were incubated under different concentration of rhBMP-2, the expression of EGFR was detected by immunocytochemistry technique, and then analyzed with HMIAS-1000 color image system. Results: rhBMP-2 had an effect on the expression of EGFR(P<0.05), which decreased in the range of 25μg/l~100μg/l rhBMP-2 and had the lowest expression at 100μg/l rhBMP-2, but up-regulated as the concentration of rhBMP-2 increased. Conclusions: rhBMP-2 has an effect on the fibroblastic phenotype of PDLC, and the appropriate dose of rhBMP-2 can assure the stability of the fibroblastic phenotype , which makes it possible to construct periodontal ligament around dental implant. ......
